In this research, diversity and population genetic structure of Castanopsis hystrix A.DC from Da Qingshan Test Forestry Farm in Guangxi Province. We using ISSR (inter-simple sequence repeat) markers in the research.The main method and results are as follows:1, The Castanopsis hystrix A. DC genome DNA was extracted with CTAB method, and the quality of the isolated DNA was high, the DNA was suitable for ISSR reaction by detection.2, The optimal ISSR reaction system suitable for Castanopsis hystrix A.DC was found, which was:20μL reaction volume, containing2.0μL10×Buffer,1.4μL Mg2+,0.3μL dNTP,0.8μL primer,0.2μL Taq DNA polymerase,50ng template DNA.Amplification program was: pre-denatrue at94℃for5min; denatrue at94℃for30s; annealing at52~55℃(temperatrue will be changing in different primer reaction) for45s; extention at72℃for2min,40cycles; last extention at72℃for7min; presevation at4℃.3, Use ISSR technology to analysis Castanopsis hystrix genetic diversity of five provenances about Da Qing Shan experimental forest in Guang xi. Choose11ISSR primers, amplified168polymorphism sites; polymorphic loci percentage is100%, the polymorphic loci percentage in different provenance between94.64%and99.40%. One of the source areas, Bo Bai has the highest percentage of polymorphic loci, as high as99.40%, it explains that the genetic variation level of Bo Bai is the highest in the five provenance area, and the Nei's genetic diversity index is0.3697, the total Shannon information index is0.550. All of the source area of the genetic similarity coefficient was between0.9503and0.9754, and the genetic differentiation coefficient between groups is0.0546, according to the groups by UPGMA cluster chart analysis shows that: Ping Xiang and Rong Xian clustering, Pu Bei and Bo Bai clustering, because of the genetic distance and geographic difference, Dong Lan, for a alone relationship, this is broadly consistent with distribution. 4> Analysis the relationship between genetic variation and geography features change of Castanopsis hystrix group on molecular level. Genetic diversity statistical analyses for32home show that that there are some differences between the genetic diversities f stanopsis hystrix in32homes. The number of effective genes, Nei's genetic diversity index, Shannon information index all reveal the richness of genetic diversity about32Castanopsis hystrix home.5, Put forward the Castanopsis hystrixof fine family protection and utilization of plan, fully understand genetic variation of Castanopsis hystrix in spatial distribution pattern to make the important role in scientific quality genetic resources protection and utilization strategy is. In genetic improvement of Castanopsis hystrix, good individual, fine group and fine provenance selection are too important to ignore. The protection of group and the individual in the group is very important, especially special habitat community and irreplaceable groups in the classification and heredity, in particular, need key protection. Such as the Bo Bai group with highest proportion have the specific genetic types which other groups do not, and the genetic diversity index of Bo Bai group is significantly higher than any other group, individual's genetic type in community is also much richer. Genetic differentiation index between all groups Gst is0.0546, genetic variation between groups is5.46%of the total variance, much lower than genetic diversity within groups. So the genetic improvement of Castanopsis hystrix should be mainly focus on the individuals, also pay attention to the group selection. Combine the group selection and good individuals, which made the most of the genetic gain.
