Orchidacea is one of the largest family of the high plants, which second only toAsteraceae. Orchids' endophytes and mycorrhizal fungi take an important part in plantecological system. It's of great significance to the in-depth understanding of Orchidacea inphylogenetic study that isolating the Cymbidium endosymbiotic fungi, to clear the diversityrelationship between Chinese Orchids and their endosymbiotic fungi.Endosymbiotic fungi from typical Cymbidium plant of Hubei province and Chongqing inChina had been isolated in the study. They had been identified preliminarily by traditionalclassification method of morphology. Further they had been identified by both rDNA ITS and18S rDNA sequence analysis of molecular biology techniques. We had studied the diversityand specificity relationship between root endosymbiotic fungi and their Orchids. Results are asfollows:1.212endosymbiotic fungi had been isolated from12Orchids of different origins,different habitats and different species. They had been preliminarily identified both bytraditional classification method of morphology and molecular biology techniques to3divisions7class12orders18families23genuses.2. In endophytic fungi, Hypocrea took obvious advantages, Gibberella,Fusarium andChaetomium were the dominant genus. Tulasnella beening isolated from C. goeringii whichgrowed in greenhouse.3. Initially revealed the diversity and specificity relationship between endosymbiotic fungifrom Cymbidium and their different origins, different species and different growth environment.Laws as follow:(1). Mycorrhizal fungi had been isolated, there were3types of Tulasnella calospora and2types of Ceratobasidium sp..The symbiotic relationship of C. goeringii and Tulasnellacalospora shows certain specificity on different places; (2). The endosymbiotic fungi beening isolated from Cymbidium except C. kanran camefrom Qingxigou showed more great diversity.(3). The diversity of endosymbiotic fungi beening isolated from C. goeringii came fromdifferent places were significantly better than that from C. faberi, C. kanran, C. ensifolium.(4). Endosymbiotic fungi separated from no matter greenhouse habitat or wild habitatboth showed strong diversity relationship; However, the distribution of the kinds of fungigenus showed large differences and the dominant fungi species separated from Orchids withdifferent habitats showed strong consistency.
