To identify the suitable range of temperature and cytokinin of Cymbidium goeringii, series of culture conditions were designed with the cross of different temperature and cytokinin (6-BA) (18℃, 22℃, 26℃, 30℃; 2mg/L, 4mg/L, 6mg/L, 8 mg/L). The activities of peroxidase (POD) and catalase (CAT), the contents of malonaldehyde (MDA), dissolved protein, polysaccharides and chlorophyl were determined by TU-1800 and other apparatuses. And we got the results below. The activities of POD show 18℃>30℃>26℃>22℃, 8mg/L>2mg/L>4mg/L> 6mg/L; the content of MDA show 30℃>22℃>18℃>26℃, 2mg/L>8mg/L> 4mg/L>6mg/L; the activities of CAT show 18℃>30℃>26℃>22℃, 8mg/L> 2mg/L>4mg/L>6mg/L; the content of dissolved protein show 26℃>30℃>22℃ >18℃, 6mg/L>8mg/L>4mg/L>2mg/L; the content of polysaccharides show 26℃ > 22℃ > 30℃ > 18℃, 6mg/L > 4mg/L > 8mg/L > 2mg/L; the content of chlorophyl show26℃>22℃>30℃>18℃, 6mg/L>4mg/L>8mg/L>2mg/L.Meanwhile, we found the plants which are cultured under 26 ℃ and 6mg/L cytokinin grow well with numerous roots, thick stems, excessive and bounty elaves. According to the data above, we know the most suitable temperature and cytokinin are 26℃ and 6mg/L. Therefore, good effection can be gained to the Cymbidium goeringi cultured under 26℃ and 6mg/L cytokinin.Separating and purifiing the polysaccharides of Cymbidium goeringii. thecrude polysaccharides were isolated from it and then being evaporated, dialyzed and precipitated with ethanol. The pure polysaccharides was obtained by two times of purification on DEAE-cellulose columns. It was identified by Sephadex G-200 gelatin column chromatography to be homogenous. Chemical analysis showed that the polymer contained uronic acid and sulfated, which account for 20.8%, 13.0% of the pure polysaccharides weight. It is proved that the main parts of polysaccharides of Cymbidium goeringii are uronic acid and sulfated groups, which accord to the previous data reported.
