Saposhnikovia devaricata (Turez.)Schichk, belongs to vivacious herbage. It is distributed mainly over Inner Mongolia, the Northeast, Hebei, and so on.There are polysaccharide, chromone, Poly acetylene, volatile oil, coumarins, organic acids and inorganic elements in Saposhnikovia divaricata(Turez.)Schichk.The polysaccharide from Saposhnikovia divaricata (Turez.)Schichk is one main active ingredient of Fang Feng, Which has the effects of anti-cancer antiinflammatory, bacteriostat, anti tumor. As so far, the research about pharmacology of Fang Feng and compound preparation is common and clinical application is widely, but the study on trace identification of ingredients is rare.In this study, the material was Fang Feng, then polysaccharide was extracted with different frequency and power ultrasonic from Fang Feng.The extraction process, activity, separation and purification, physicochemical property, structure, molecular modification, the affect to structure from ultrasonic were studied.The results were as follows.1. Ultrasonic-assisted extraction under3different power and4frequency was adopted to extract polysaccharide from Saposhnikovia divaricala (Turez.)Schichk, the antioxidation experiments were done in12kinds of polysaccharide obtained.The result indicated that the maximum recovery rate was7.12%under135kHz,290W.The antioxidative activity was higher when the ultrasonic frequency was80kHz or135kHz and ultrasonic power was in the range from150W to220W. The extraction rate of S.divaricata polysaccharides can be improved by ultrasonic extraction, proper ultrasonic power and frequency can reach a better antioxidation activity effect.2. Saposhnikovia divaricata(Turez.)Schichk extracted under12different frequency and power was separated and purificated,19kinds of pure polysaccharides were obtained. HPLC and IR were used to reach a conclusion that as the power of ultrasonic began to increase, molecular weight of Saposhnikovia divaricata(Turez.)Schichk was larger, as the frequency began to increase, molecular weight of Saposhnikovia divaricata(Turez.)Schichk was smaller, ultrasonic produced some effcets on Fang Feng, but the features of polysaccharide was not changed.3. Saposhnikovia divaricata(Turez.)Schichk(U-SPS) extracted under80kHz and 220W, U-SPSO and U-SPS1were obtained by separation and purification, UV-spectrophotometer, ESEM, AFM and Thermal analysis, were used to study the two different polysaccharides. The result indicated that, SPSO and U-SPS1, uronic acids were12.07%and0.95%, U-SPSO was dissolved in water more easily, ESEM and AFM showed that U-SPSO was neat and orderly, a chain and many branches structre. but U-SPS1was opposite and contained a few branches, U-SPSO's heat stability was better between26℃and800℃.4. HPLC, GC, IR, periodate oxidation and Smith degradation, NMR were used to determine structural analysis of U-SPSO and U-SPS1. The results:relative molecular weight of U-SPSO and U-SPS1were:2.78×104Da,7.94×104Da; Both U-SPSO and U-SPS1contained Ara, Man, Glc, Rhaand Gal.The configuration of U-SPSO is β configuration, The structural repeating unit of polysaccharide chain may be:The configuration of U-SPS1is β configuration, The structural repeating unit of polysaccharide chain may be:5. Further, purified Cordyceps militaris polysaccharides were successfully modified by either sulfation, carboxymethylation or acetylation and their antioxidant activity and that of their modification products were comparatively assessed based on the ability to in vitro scavenge superoxide anion, hydroxyl and alkyl free radicals.
