Camptotheca acuminata Decaisne which belongs to genus Camptotheca, Nyssaceae family is25-30meters high. C. acuminate is the only well known species of its genus and it is distributed in south-eastern china mostly along the Yangtze River, growing in forests and on hillsides at altitudes between250and1000metres. The genus C.acuminata is cultivated as an greening trees, but rencently its greatest commercial value lies in its secondary metabolites production-camptothecin(CPT) and10-hydroxycamptothecin(HCPT), which have emerged as one of most promising agents for cancer treatment because of their inhibitory activity against tumour cells. The paper has a comprehensive and thorough research from two sides of the-inductive rate of adventitious buds, with explants from stems and leaves of camptotheca acuminate. Adventitious buds induction, callus induction, proliferation and regenerative potentialities of calli from Camptotheca acuminata under different light qualities. These results provide fundamental basis for the further research of genetic transformation, cell culture, plant regeneration and secondary metabolites acquisition in Camptotheca acuminata. The result is as following:1. Among the MS,1/2MS. WPM and SH four basic mediums under3kinds of light qualities(red,blue,white),the SH is the best media for the stem and leaf callus induction supplemented with0.075mg·L-1TDZ and1.0mg·L-1NAA under the blue light. The callus without browning, is green compact nodule.2. There is a big difference in the inductong of adventitious buds for leaves and stems.The induction rate of stems was higher than the induction rate of stems. Auxin contrast test was done, which NAA is better than IAA for stems and IAA is better than NAA for leaves.The best condition for stem:SH+1.0mg·L-16-BA+0.05mg·L-11NAA(blue light) and best condition for leaves:SH+2.0mg·L1-6-BA+0.1mg·L-1IAA(white light), induction rates were24.1%and18.3%.3. Regenerative potentialities of calli of Camptotheca acuminata:(1) At the7th day of subculture under blue light, the calli had the highest SOD activity and then it had a decreasing tendency after the7th day.There was always had low SOD activity under the red light.(2) At the7th day of subculture under blue light, the calli had the lowest POD activity which was1/3POD activity under the white. From7th to14th, the POD activity had a obviously enhance and had the highest POD activity at the14th under the blue light.The change of SOD activity is contrary to the change of POD activity under the blue light which was subculture on the SH medium.At the7th day of subculture under blue light, the calli had the highest SOD activity and the lowest POD activity, which might suggest a highly potential of regeneration of them.
