Study On Genetic Diversity Analysis Of Camptotheca Acuminate Provenance Markers

The seed of seven Camptotheca acuminate Provenance was collected fromJiang province,Guizhou province,Yuannan province,Guangdong province and Hubei province.,and the seed morphological characteristics,some seed components,seedling growth,Camptothecin content of leaves and gene diversity index were measured.The main results were as follows:1.There were significant differences in the biological characteristics of tree seeds in different provenance.The range of seeds length,width and height were 19.69~24.20 cm,5.37~6.82 cm and 3.09~3.67 cm,and the range of thousand seed weight and the germination rate were 28.93~48.49 g and 46.5%~79.87%.The range of soluble sugar and camptothecin content of seed were 4.23~9.16mg/g and 1.27~1.74mg/g.The seed from Yunnan province had highest camptothecin content,and it was good materials used for extracting camptothecin.2.The seedling growth of different provenance had significant difference.The range of seeding height and ground diameter were 15~126cm and 6.49~14.57 mm,and the range of main root length were 13~53cm.The range of chlorophyll content,soluble sugar content and soluble protein content in seedling leaves were 1.005~1.552mg/g,50.94~205.46mg/g and 37.79~97.82mg/g respectively.The range of seedling above-ground biomass and under-ground biomass were 1.01~34.09 g and 0.87~12.57 g.3.The provenances from Libo of Guizhou province and Nanjing of Jiangsu had best growth characteristics,while the provenance from Xingang of Guangdong province had highest camptothecin content and yield in leaves,and its content and yield were 0.664mg/g and 3.420mg/tree.4.The DNA was extracted from the seedling.Using the method of ISSR markers,six primers were selected from 100 primers to classify the tree.The results showed that six ISSR primers had been expanded to 433 sites.Each primer had 58 to 92 pieces of DNA,and the average was 72 pieces.There were 377 diversity sites,and polymorphism percentage was 87.1%.The primer ISSR36 and ISSR41 surpassed 90% was,and the polymorphism of the primer ISSR36 reached 97.1%.DNA band polymorphism could distinguish the different provenance.
