With awareness of environmental protection, the chemical pesticides which highly toxic.high residue will be limited gradually, and the Bio-pesticides which on pest efficient, non-target organisms security.environment friendly will be easy to decompose gradually by the publicrecognition. The main way to discovery a new Bio-pesticides is looking for the compounds that agricultural activity from nature or searching the active lead compounds from natural products then synthesize or modify their structures.For this purpose,this experiment studies on the insecticidal antibacterial activity of Rutaceae Murraya paniculata L.and separate the chemical composition,structure identification of Murraya paniculata L.The results are as follows:1.The extraction.separation and identification of the active ingredient.Using methanol percolation method to extract the active substances of Murraya paniculata L.,followed by with petroleum ether,chloroform,ethyl acetate,n-butanol.Than chloroform extract by thin-layer chromatography, silica gel column chromatography, gel column chromatography separation and purification,two compounds were getted. By1HNRM,13CNRM and MS identification,the compound I was3,5,6,7,3',4',5'-seven methoxyflavone,the compound Ⅱ was3,5,7,8,3',4',5'-seven methoxyflavone.2.The determination of insecticidal activities.Using the methods of bioassay and chemical separation to combine.Muscadomestica,Predenia liture.Kaltenbach indicated,Brontispa longissima Gestro for the test target standard pest, and tested the insecticidal activity on four kinds of extraction material of Murraya paniculata L.,and the further separationthe composition of the insecticidal. The results showed that:(1)When the pharmaceutical treatment after48h at the concentration of5mg/mL,the chloroform extract toxic to Muscadomestica mortality rate of91.67%;(2)When the pharmaceutical treatment after72h at the concentration of8mg/mL,the chloroform extract on the cytotoxicity of Lipaphis erysimi(Kaltenbach) mortality rate reached74.00%;(3)When the pharmaceutical treatment after48h at the concentration of10mg/mL.the cntactactivity of the chloroform extract against the3rd and5rd instar Predenia liture,the mortality rates were72.22%,64.15%;(4) When the pharmaceutical treatment after48h at the concentration of10mg/mL, he cntactactivity of the chloroform extract against the3rd and5rd instar Predenia liture,the mortality rate was more than60%.The insecticidal activity of the chloroform extract is the best.Continue after their chromatographic separation and petroleum ether chromatography and Ll-L5.The L3showed good insecticidal activity,the pharmaceutical after12h,the housefly mortality rate was more than90%. Further separation of L3components,getted the compound I and compound II. Found that the compound I significant insecticidal activity.the LC50of Muscadomestica was0.1147mg/mL; the LC50of Lipaphis erysimi(Kaltenbach) was0.1201mg/mL3.The determination of antibacterial constituents.Using the growth rate method, Colletotrichum musae, Colletotrichum gloeosporioides, Fusariun oxysporum, Fusarium oxysporum,Botryodiplodia theobroma these five plant pathogens as a target bacteria were tested paniculata petroleum ether, chloroform, ethyl acetate, butanol extract and supercritical CO2extraction method of extraction of essential oils antimicrobial activity tests. The results show that, when the concentration was0.5mg/mL, the chloroform extract inhibit the effect of the tested strains, the inhibition rate of Colletotrichum musae (Berk&Curt) Arx and Colletotrichum gloeosporioides Penz were the best,they were68.10%and63.52%.respectively;the essential oils against banana anthrax and mango anthrax, the inhibition rates were67.80%and61.02%; Other extracts of the inhibitory effect of the pathogen is not ideal. Chloroform extract and deal with other solvent compared antibacterial rate reached a significant level.The Virulence test showed that,the essential oils against Colletotrichum musae (Berk&Curt) Arx.the EC50was0.2611mg/mL; the component26against Colletotrichum musae (Berk&Curt) Arx and Colletotrichum gloeosporioides Penz,the EC50was0.0786mg/mL and0.0915mg/mL.4.The study of insecticidal mechanism.With Muscadomestica and Lipaphis erysimi(Kaltenbach) for insects,to explore on the affect of tthe compounds I against the acetylcholinesterase, carboxylesterase, glutathione-S-transferase enzyme of Muscadomestica and Lipaphis erysimi(Kaltenbach). The results show that,the processing insects reduced acetylcholine content than in the control of insects, suggesting that the compound I insecticidal mechanism of synaptic neurotransmitter release is related to inhibition of acetylcholinesterase activity on Muscadomestica and Lipaphis erysimi(Kaltenbach).The same time, the compound I carboxylesterase of the Muscadomestica and Lipaphis erysimi(Kaltenbach), glutathione-S-transferase a certain extent.lt is the preliminary judging, the compound on the inhibition of acetylcholinesterase, the cumulative poisoning as acetylcholine, while inhibiting the enzymes detoxifying enzymes carboxylesterase and glutathione-S-transferase, resulting in detoxification enzymes can not play a role,and ultimately cause the insects to death.5.The essential oils and components of26analysis by GC-MS By gas chromatography-mass spectrometry (GC-MS) to analyze the chemical composition of The essential oils and components of26.The essential oils which Supercritical CO2extract was identified20compounds, accounting for99.99%of the total essential oil. The main components were n-hexadecanoic acid (28.76%),1-(1,5-dimethyl-4-cyclohexene-yl)-4-methylbenzene(22.38%),N-(1-methyl-3-O-butylene)-4-methoxyani line (20.11%).The main component of the component26for the7-methoxy-8-prenyl coumarm.
