ABA is an endogenous regulatory factor in plant growth and development,which is asimportant, even more important role in fruit ripening and softening.Ethylene is the earliestfound one of plant hormone, research shows that play a role in the plant seed germination,promoting flowering, flowers and leaves fall off and fruit mature aging softening andphysiological processes. In this paper make the BaiLi peach feuit as test materials, its sizeuniform maturity consistent mechanical damage to fruit with exogenous ABA treatmentrespectively, ABA inhibitors (NDGA) processing to block the endogenous ABA synthesisand CK water as a comparison, determined the fruit hardness, respiratory rate, ethylenerelease rate,key enzymes of ethylene biosynthesis ACC synthase and ACC oxidase activityand cell walls degradation related enzymes PME and PG enzymatic activity to konw theinfluence of ABA on fruit ripening and ethylene biosynthesis.This test also use RT-PCRmolecular biology technology in ethylene signal transduction pathways genes and ethylenereceptor gene expression quantity analysis.The experimental results are as follows:1, peach fruit postharvest storage period hardness down fast, ABA treatment can improvecell walls enzyme degradation PG and PME enzymes activity, exogenous ABA treatment canaccelerate softening process, but instead, with NDGA processing will slow down thesoftening rate.2, the ABA treatment can improve ethylene biosynthesis key enzyme ACC synthase andACC oxidase activity, exogenous ABA can make the ACC synthase and ACC oxidaseactivity peak appeared in advance, and are accompanied by a peak of the early release ofethylene appear.3, the RT-PCR show that exogenous ABA treatment can improve ethylene receptor geneexpression of ETR1quantity, exogenous ABA treatment can reduce ethylene signaltransduction pathways gene expression of CTR1quantity, can improve the ethylene signaltransduction pathways gene EIN2, EIL1, EIL2expression volume.
