Chocolate spot is an important disease in broad bean production caused byBotrytis spp. pathogens Botrytis cinerea, Botrytis fabae, Botrytis fabiopsis. Itoccurred in China and the world. When the climate is suitable, this disease occurseriously. Botrytis spp.isolates separated from Gansu, Qinghai, Jiangsu, Anhui,Sichuan, Hubei Hebei, Chongqing were identified in morphology and molecularbiology techniques in this study and do sporulation experiments and the analysis ofgenetic diversity. Besides, we screend161broad bean varieties,the result as follows:1. In the year2010~2011, we separated choaolate spot samples from8provinces, three kind of pathogens isoalates were identified by morphology andmolecular biology techniques. A total of314isolates were identified, containing84Botrytis fabae isolates,110Botrytis cinerea isolates,120Botrytis fabiopsis isolates.314isolates were detected by virulence,all the isolates produce different size of lesionafter3days.2.110Botrytis cinerea pathogen iaolates were analyzed using technique ofmorphology observation, Flipper and Boty transposons detection, Bc-hch sequenceamplification and restriction, Simple sequence repeat(SSR).110strains of Botrytiscinerea isolates show mycelium, conidial, sclerotia three types in culturalcharacteristics and the main type is sclerotia.110isolates are divided into threecluster groups at Similarity coefficient0.702and show the genetic differentiation hascontact with geographic environment in Botrytis cinerea strains.3. Botrytis cinerea, Botrytis fabae, Botrytis fabiopsis isolates were separated50respectively and analysis the genetic diversity, A total of123clear and pepeatedbands were amplified Using tenth polymorphic ISSR primer pairs in150Botrytis spp.isolates, gene diversity vary from3.7770~6.6320, Shannon's information index varyfrom5.7400~9.1100, the increasing gene diversity and Shannon's information indexconnect with respective increasing sample number.4. we chose Botrytis cinerea, Botrytis fabae, Botrytis fabiopsis isolates forsporulation experiment, establishing a new method of prouucing conidias. Aslo,31 resistant varieties were identified in161broad bean cultivars.
