Objective:To discuss the difference of integrinβ1 and calcium- and integrin-binding protein 1 (CIB1) expression in the renal tissues between renal cell carcinoma and nomal renal tissues,to investigate the correlation between them and the clinicopathological parameters,and to provide theoretical basis for their function and role in the tumorigenesis,proliferation,invasion and metastasis of renal cell carcinoma.Methods:We randomly selected tumor samples from 62 surgically removed kidneys containing RCC,20 samples of normal renal tissues as controls. All patients underwent nephrectomy in the department of Urology in the second hospital of Tianjin Medical University from January 2007 to January 2009.All tumor samples are proved to be RCC by pathology. integrinβ1 and calcium- and integrin-binding protein 1 (CIB1) expression was investigated by an indirect immunoperoxidase technique(S-P method of Immunohistochemistry) in the tissues of RCC and normal renal.Results:The positive expression rate of Integrinβ1 in RCC (56.6%) was significantly higher than that in normal renal tissues (20.0%) (P<0.05). The positive expression rate of Integrinβ1 showed a significant relation with TNM stage,histological grade,tumor size,incidental renal carcinomas or not and lymph node metastasis in RCC. The expression of Integrinβ1 in late clinical stage (ⅢtoⅣ) (81.8%) was significantly higher than that in primary clinical stage(50.0%).The expression of Integrin(31 in low histological grades was distinctly lower than that in high histological grades(P<0.05), but the difference between low histological grades and second histological grades, high histological grades and second histological grades was not significant(P>0.05).The difference of Integrinpβ1 expression positive percentage was signifigent between diameter of tumor less or more than 3cm,(P=0.007).The expression of Integrinβ1 with lymph metastasis(88.9%)was statisticaly higher than that without lymph metastasis(50.9%),(P=0.034).The abnormal expression rate of CIB1 in RCC (61.3%) were significantly higher than normal renal tissues(35.0%),(P<0.05).The positive expression rate of CIB1 showed a significant relation with histological grade and lymph node metastasis in RCC.The difference of expression of CIB1 between different histological grades was not significant. The positive expression rate of CIB1 with lymph metastasis(100%) was significantly higher than that without lymph metastasis (54.7%),(P=0.01).There was a significant positive relationship between expression of CIB1 and Integrinβ1 in RCC.(r=0.4501, P<0.05).Conclusions: The positive expression rate of Integrinβ1 in RCC was much higer than normal renal tissues. Integrinβ1 might be valuable markers in assessing biological behavior, invasion and metastatic potential and prognosis in RCC.Compared to normal renal tissues,the expression of CIB1 rised in RCC. Abnormal expression rate of CIB1 was significant relation with TNM stage,tumor size and lymph node metastasis.The expression of CIB1 might be used as an objective indicator of the metastatic potency of local lymph node, extent of invasion and the prognosis in RCC. Positive correlation was found between low expression of CIB1 and high expression of Integrinβ1 in RCC. Evaluating both of their positive rates might be useful to tumor diagnosis,estimating potency of invasion and metastasis, evaluating the prognosis of RCC and guiding the reasonable comprehensive treatment.
