Abstract/Summary:
A large number of epidemiological, clinical and laboratory study demonstrated that the integration of HBV virus has tightly connection with the hepatocellular carcinoma(HCC). We need to find the inserted site of the HBV in the human genome by the Alu-PCR, to Find the inserted site of the HBV need the further study because we have not acquired the completed the inserted fragment.We found the difference of Meprin gene in the HCC samples by the screening of the genetic chip. Through the semi-quantitative RT-PCR, the further study in 47 cases of cancer and adjacent tissues found that the expression of Meprin-αsignificantly increased in 10 of cancer tissues, whereas the expression of Meprin-βsignificantly increased in 22 of adjacent tissues. The Stable expression of Mepla and Meplb in cell lines shows mepla significantly promote the growth of Focus cell line, and mepla and meplb significantly promote the migration of SK and Focus cell line, but we still need further verification; Transfected with Huh7 cell lines by the p-SUPER vector, we found that the growth of Huh7 cell lines was significantly inhibited for the interference expression of mepla. We investigated the relation between the Plasminogen activator system and Meprin gene, and found there is no obvious connection in human hepatic carcinoma cell.
