Objective To study the effects of Helicobacter pylori in gastric carcinoma lymph node metastasis in vitro and in vivo.Methods Immunohistochemistry technique was used to detect the expressions of D2-40 in 80 gastric carcinoma specimens and to count the lymphatic vessel density, Hp infection was determined by improved Giemsa method and rapid urease test. The results were analysed for the relations among Hp infection and gastric carcinoma lymph node metastasis and the LVD of gastric carcinoma and their e clinical pathological characteristies;The Hp strain SSI was culture in 5%O2,10%CO2,85%N2 and 37℃. Different concentrations(104,105,106,107cfu/ml) of Hp Co-culture with human gastric cancer cell line SGC-7901 for 6,12,24hour. Immunohistochemistry technique was used to detect the expressions of VEGF-C in SGC-7901, The results were analysed for the relations among Hp and VEGF-C in different concentrations and different Co-culture time.Results 1 The infection rates of Hp in gastric carcinoma group was 66.25%. Hp infection was correlated with lymph node metastasis and clinical TNM stage but irrelevant to sex, age and pathology histological.2 The Hp positive group LVD(16.33±6.89) was higher than the Hp negative group LVD (9.10±3.10), (P<0.05) the differences had significant. LVD in gastric carcinoma specimens was correlated with lymph node metastasis and clinical TNM stage but irrelevant to sex, age and pathology histological.3. To streak inoculation for 24 hour the Hp colony were scanty and like needle tip. When cultured for 72 hour Hp colony were round, similarity round, slick evection borderline regularity and semitransparent. When cultured more than 72 hour the Hp colony were not become more but Hp can become globular. Hp was Gram-negative bacterium,comma,benting like S or short rod and rapid urease test, oxidation enzyme test and hydrogen peroxidase test were positive.4 Immunohistochemistry result:Expression of VEGF-C protein In logarithmic phase gastric carcinoma cell the expression of VEGF-C protein positive rate is 61.98±0.71%,but the cell culture longer the expression of VEGF-C protein positive rate is not more, the differences had no significant (P> 0.05). Different concentrations of Hp Co-culture with gastric carcinoma cell longer time the expression of VEGF-C protein positive rate is more, the differences had significant (P <0.05) The same Co-culture time high concentrations of Hp the expression of VEGF-C protein positive rate is more, the differences had significant (P<0.05)Conclusions 1 In gastric carcinoma tissue the lymph vessel mostly in the borderline, the lumens were irregularity, partly had carcinoma cell in them, and associated with the lymph node metastasis, Hp infection in lymph node metastasis group LVD higher than without lymph node metastasis group LVD. Lymph node metastasis and LVD was direct correlation. LVD and the rate of lymph node metastasis in Hp positive group was higher than the Hp negative group. Hp may step up LVD of gastric carcinoma tissue, Hp infection may promote the lymph node metastasis in gastric carcinoma.2 Hp requirement very severity in culture medium and culture environment. It Severity confine the clinic and foundation study of Hp. The penetrating study of Hp culture has pragmatic value in.pathopoiesis,Hp drug resistance and choice antibiotic to eradicate Hp.3 In vitro experiment without of the complicated inflammatory medium and cytokine in vivo.In this study Hp can promote the expression of VEGF-C protein in gastric carcinoma cell SGC-7901 and associated with the Co-culture time, Hp concentrations.So we presume that Hp can promote the expression of VEGF-C protein accordingly promote the lymph node metastasis in gastric carcinoma.In early time eradicate Hp may decrease the lymph node metastasis of gastric carcinoma.
