Expression Of VEGF-A, D And Podoplanin In Advanced Gastric Carcinoma And Its Relationship With Lymph Node Metastasis

To study the relationship between the vascular endothelial growth factor A , D (VEGF-A,D) and the lymphatic metastasis in advanced gastric cancer , To observe the expression of podoplanin in lymphatic endothelial cells in the gastric carcinoma so as to provide morphological data for the study on the molecular mechanism of tumor metastasis. Method: Immunohistochemical SP method was used to examine the expressions of the VEGF-A and D in 54 advanced gastric cancer tissues , 54 pericancerous gastric tissues ,12 normal gastric tissues, and the expression of podoplanin in gastric carcinoma and normal gastric tissue;and examine the expression of three factors in two groups (positive and negative for lymph node metastasis) ,Using podoplanin as the specific marker of lymphatic endothelium,lymphatic micro-vessel density(LMVD) in advanced gastric cancer tissue and pericancerous gastric tissue was calculated. Results: The positive expression rate of VEGF-A and D in advanced gastric cancer tissue and pericancerous gastric tissue was significantly higher than that in normal gastric tissues (P<0. 01), in positive lymph node group the expression rate of two factors was obviously higher than negative group (P<0. 05), The expression of VEGF-A and D in gastric carcinoma was positively correlated with lymph node metastasis(P<0. 01). Podoplanin was positively expressed in the gastric carcinoma lymphatics. LMVD detected from the VEGF-A,D positive group respectively was significantly higher than that in negative group , LMVD detected from positive lymph node group was obviously higher than negative group. Conclusion: It suggests that VEGF-A and VEGF-D play an important role in lymph node metastasis of advanced gastric cancer,both factors increase LMVD detected from advanced gastric cancer tissues and pericancerous gastric tissues collaboratively.The specificity that podoplanin marker lymphatic is stronger.
