Purpose:Cordyceps Sinensisi (Cordyceps Sinensisi (Berk.) Sacc) is a rare traditional Chinese medicine in China, with lungs and kidney, the efficacy of phlegm to stop bleeding, can be used to treat chronic cough virtual asthma, labor cough hemoptysis, impotence and nocturnal emission, waist and knee pain, etc. , but the shortage of resources, expensive. Most are available in the market by deep fermentation of Cordyceps sinensis mycelium as an alternative to natural Cordyceps. We choose to organize the wild Cordyceps stroma separated to obtain pure asexual stage, the caterpillar fungus (code 832) strains. Optimize the fermentation medium and fermentation process, a preliminary study of its pharmacological effects, and fingerprint analysis used to control its intrinsic quality, has great significance, for the Cordyceps sinensis (code 832) mycelium in the further development and research to provide the necessary the basic work.Material and method:We choose to organize the wild Cordyceps stroma separated to obtain pure asexual stage, the caterpillar fungus (code 832) strains. In this study, dry weight of mycelium, extracellular polysaccharide, adenosine mycelium of indicators for the test, by single factor test and orthogonal test of Cordyceps sinensis (code 832) strains of deep fermentation medium and fermentation conditions, Cordyceps mycelium were mouse anti-fatigue test, hypoxia test and anti-inflammatory tests in rats, a preliminary study of its pharmacological effects, and finally HPLC fingerprint method for quality of Cordyceps sinensis mycelium provide reference basis for evaluation.Conclusion:1 To Cordyceps (Cordyceps Sinensisi (Berk.) Sacc) code 832 strain as the material, by single factor test and orthogonal experiment with glucose as the carbon source, beef extract as nitrogen source, potassium dihydrogen phosphate, magnesium sulfate as the inorganic Cordyceps sinensis is more suitable when (code 832) strains of submerged fermentation, and the best medium for glucose, 6.5 g / L, beef extract 6.5 g / L, potassium dihydrogen phosphate 1.7 g / L, magnesium sulfate 1.5 g / L . Through the optimization of the fermentation process is obtained when the inoculum was 15% in 250 mL of flask are installed with liquid 75 mL, initial pH 7.0, and the temperature at 18℃under cultivation, the most suitable for Cordyceps sinensis (code 832) strain fermentation. The optimized fermentation medium and fermentation conditions, can yield a substantial increase in fermentation products, higher than the national pharmacopoeia standards.2 Through a number of pharmacological tests, and the same research group compared the results of preliminary tests show that Cordyceps (code 832) mycelium can very significantly improve the mice's physical strength, to extend their swimming time, and after exercise can reduce the serum in mice urea nitrogen, blood lactic acid, increased serum liver glycogen and muscle glycogen content, with a clear anti-fatigue effect; Cordyceps sinensis mycelium can also prolong the time of hypoxia tolerance in mice; can significantly improve the symptoms of arthritis in rats by reducing serum content of TNF-α, thereby inhibiting the proinflammatory effects of cytokines.3 By Cordyceps (code 832) mycelium of the HPLC fingerprint was established to Agilent Eclipse XDB-C18 column (4.6 mm×150 mm, 5μm) column was used; mobile phase of 5% to 60% methanol gradient elution; flow rate was 1.0 mL / min; column temperature of 30℃; detection wavelength 260 nm; injection volume was 10μL, all components detected within 40 min in the complete method of analysis. The method validation of the method precision, repeatability and accuracy are good, you can better control the quality of Cordyceps sinensis mycelium...
