Inonotus obliquus is a rare medicinal fungi. Polysaccharide is one of the most important active ingredients of Inonotus obliquus. It has excellent effects at the aspects of anti-cancer, reducing blood sugar, anti-virus, reducing blood fat, enhancing immunity,antioxidation,anti-thrombosis,anti-inflammatory,assuagingpain,etc.Currentl y resources of wild Inonotus obliquus gradually are limited, and the technique of artificial cultivation has not been developed yet. Obtaining polysaccharide by fermentation has become extremely important.In this paper, we selected a strain for fermentation, optimized the deep fermentation cultivation conditions to achieve the objective of high-yield poly-saccharide production and studied the anti-fatigue effects of the polysaccharide of Inonotus obliquus on mice. The main results were as follows:1. Using strain No.l and No.2 as materials, we compared their mycelial growth rate by plate culture, mycelium growth vigour and yield of crude polysaccharide by deep fermentation cultivation. The hypha growth rate of strain No.l is 1.872 times than strain No.2.The mycelia dry weight of strain No.1 is 1.254 times than strain No.2.The polysaccharide yield of is 1.167 times than strain No.2. And the strain No.1 is hard to become brown or age.No.1 strain is suitable for the deep liquid fermentation.2. We carried out the liquid fermentation experiment by combination of single-factor test and orthogonal test. Taking yield of crude polysaccharide as major evaluation factor, we investigated the effects of polysaccharide production by different culture media composition and fermentation conditions systematically. The optimal medium ingredient is:corn flour 20g/L,glucose 15g/L,yeast powder 3g/L,bran 30g/L,KH2PO4 lg/L, MgSO4 · 7H2O 0.5g/L,VB1 0.005g/L;The optimal fermentation condition for polysaccharide production were:temperature 30℃, rotational speed 140r/min, initial pH 6,fermentation time 12d,inoculation quantity 12.5%, seed age 7d.The yield of crude polysaccharide reached 0.511g/100mL.3. By setting up mouse fatigue models, we carried out the research on the anti-fatigue effects of the Inonotus obliquus polysaccharide from fermentation liquid. The weight of mice in experiment groups didn’t change significantly compared with the normal control group. The swimming time of IO-P3 high-dosage group was improved by 52.8% significantly compared to the control group in swimming time measurement (P<0.05). The serum urea nitrogen level of mice of IO-P3 high dosage group reduced 31.3% than the control group significantly in serum urea nitrogen measurement (P<0.05).The area under the acid curve of mice blood lactic deceased 41.3% significantly compared with the control group in blood lactic measurement (P<0.05). The concentration of hepatic glycogen of IO-P3 high dosage group was improved by 30.6% significantly compared to the control group in hepatic glycogen measurement (P<0.05). These results suggest that the polypeptide of Inonotus obliquus could alleviate fatigue of the mice significantly. No difference was observed between all dose groups of intracellular polysaccharide IO-P1 from culture medium no constituent of the birch wood chips and intracellular polysaccharide IO-P3 from culture medium which contain birch wood chips(P>0.05). And no difference was observed between all dose groups of extracellular polysaccharide IO-P2 from culture medium no constituent of the birch wood chips and extracellular polysaccharide IO-P4 from culture medium which contain birch wood chips(P>0.05).
