Object:To confirm the effectiveness of Minkang oral solution in treating allergic rhinitis and find out the mechanism underlying.Method:1 Prepare rat model of AR: Mixed OVA, Al(OH)3, and bordetella pertussis together in normal saline. Rats were allergized with this suspension as follows:Injected suspension into the four foot pads on the first day. Five days later they were boosted with injection of physiology saline containing OVA into the rostral parts of the back. And fourteen days later, sensitization was performed every day by dripping OVA in saline into thebilateral nasal cavities.2 Dose-effect relationship of Minkang oral solution: Using rat as model animal, dividing them into different groups and modeling, each of the five medication administration group was intragastrically administrated with Minkang oral solution in different doses (5g/kg, 10g/kg,20g/kg,40g/kg and 80g/kg). The severity of AR was assessed by the content of EOS and HA in peripheral blood by SYSMEX and fluorospectrophotometer, combining the result of pathological section of nasal mucosal.3 Dose-time relationship of Minkang oral solution: Using rat as model animal, dividing them into different groups and modeling, each of the five medication administration group was intragastrically administrated with Minkang oral solution in different duration (3days,5days,7days, 10days and 14days). The severity of AR was assessed by the content of EOS and HA in peripheral blood by SYSMEX and fluorospectrophotometer, combining the result of pathological section of nasal mucosal.4 Influence in the stabilization of membrane of RMPC: Using rat as model animal, and extracting RMPC after immunized, cells of medication administration groups were cultured in buffer A, containing different concentration of aseptic Minkang oral solution (0.216g/mL,0.432 g/mL, and 0.864g/mL). The result was observed via inverted microscope.Result:1 Dose-effect relationship of Minkang oral solution: The dose-effect relationship was confirmed by assessing the content of EOS and HA in peripheral blood. In the dose rank of this experiment, the content of EOS and HA decreased according to the increasing of dosage. Comparing to the blank control group,content of EOS and HA were obviously higher in model group (P<0.01),and section of nasal mucosal revealed sever pathological changes;on the contrast, content of EOS and HA were all obviously decreased in medication administration groups contrasting with model group(P<0.01),and pathological changes of nasal mucosal were ameliorated.2 Dose-time relationship of Minkang oral solution: The dose-time relationship was confirmed by assessing the content of EOS and HA in peripheral blood.In the dose rank of this experiment,the content of EOS and HA decreased according to the increasing of dosage.Comparing to the blank control group, content of EOS and HA were obviously higher in model group (P<0.01),and section of nasal mucosal revealed sever pathological changes;on the contrast, content of EOS and HA were all obviously decreased in medication administration groups contrasting with model group(P<0.01),and pathological changes of nasal mucosal were ameliorated.3 Influence in the stabilization of membrane of RMPC: The rate of positive cell in model group was obviously higher than that in blank control group (P<0.01),and the rate in medication administration groups were obviously lower than that in model group(P<0.01).Conclusion:1 Minkang oral solution is effective in treating allergic rhinitis. 2 The minimal effective dose of Minkang oral solution in healing allergic rhinitis is 5g/kg;and the maximal effective dose is 80g/kg.3 Minkang oral solution began to take effect in 3 days,and it would approach the maximal effect in 14 days.4 Minkang oral solution is able to ameliorate pathology changes in nasal mucosal.5 Minkang oral solution is able to stabilize the membrane of RMPC.
