Flos Magnoliae is one of commonly used Chinese herb which belongs to the category of relieving Exterior syndrome. It has the function of expelling Cold and clearing the nasal passage. It is frequently used for treating the disease of otorhinolaryngology especial allergic rhinitis. We had researched the anti- inflammatory effect and mechanism of volatile oil. And our laboratory has previously demonstrated that the volatile oil is one of the major effective components of this herb, and its anti-hypersensitive function is better than that of water-solubility ingredient. But there are few pharmacological researches about its anti-hypersensitive function, let alone its mechanism. This dissertation firstly studied the anti-hypersensitive effect mechanism of volatile oil of Flos Magnoliae (VOMbP), from the level of entire animal, cell and molecule thoroughly and systemically and systemically, by means of advanced modern pharmacological methods such as cell culture, ELISA, Immunohistochemistry , analysis etc . The results of the experiments showed that:1. The therapeutical effect of VOMbP at the allergic rhinitis(AR) was significant and clear-cut. VOMbP is one of the primary substantial basic of its anti-hypersensitive function. The therapeutical effect of VOMbP were as follows: (l)inhibitingthe increase in nasal allergic symptoms; (2)relieving the degree of the nasal mucosal engorgement and edema, (3) reducing the increased number and infiltration of mast cell and eosinophil.2. The cellular and molecular pharmacological mechanisms of VOMbP at AR were as follows: (1) Stopping the occurring and development of AR when it start by preventing the increase of Th2 factor as IL-4 in hypersensitive guinea-pig body ?consequently regulating the maladjustment of Thl/Th2 cell; (2) inhabiting eosinophil transude from vessel by restraining the expression of ICAM-1 in Endothelial cell, then controlling the adhension between Endothelial cell and eosinophil.3. The anti-hypersensitive function of VOMbP is irrelevant to the release of O2 from eosinophil .4. Mending the way of developing AR model and the way of checking cell adhesion . HRP was used to study the influence of VOMbP on eosinophil-endothelium adhesion. We developed a model of nasal allergy of rat by using nose drop of 10% TDI for 14 days. The results of experiments indicated that our betterment is feasible, simple and convenient.5. The results of experiments demonstrated that there were no significance increases in the expression of VCAM-1 in the epithelium.
