Anthriscus sylvestris,the representative plant of Anthriscus sylvestris (Linn.) Hoffm,are used with its root and leaf.As are the special local products in Emei mountain,Anthriscus sylvestris are frequently used as Chinese herbal medicine and tonic food of certainly curative effect.In this dissertation,we engage in studies on acute toxicity of anthriscus sylvestris,the best extraction condition of flavonoids in anthriscus sylvestris and its antioxidant activity,the antioxidant activity and immunomodulatory effect of polysaccharide in anthriscus sylvestris.(1) We exam the acute toxicity of water decoction and petroleum ether extract of anthriscus sylvestris,through preliminary experiment and maximally tolerated dose experiment.The results show that Anthriscus decoction and the petroleum ether fraction of the mice were equivalent to the maximum tolerated dose of adult body weight of 60kg 15g daily dosage of 200 times and 1,000 times,all with security.(2) The orthogonal design are used to investigate the best extraction condition of ultrasonic extraction of flavonoids in anthriscus sylvestris,to L9(34)arranged orthogonal test.The optimal extraction conditions were:ultrasonic time 25min,ethanol concentration 60%,the material fluid ratio is 1:15.(3) Antioxidant properties of flavonoids in anthriscus sylvestris are measured in vitro through Fenton reaction and pyrogallol autoxidation method.The result showed that flavonoids in anthriscus sylvestris had a strong capacity on scavenging hydroxyl free radical and superoxide anion freeradical.With the increase of concentration,antioxidant activity increased.The scavenging rate of hydroxyl free radical is 32.88%,the inhibition rate of superoxide anion free radical is 24.87% when the flavonoids concentration was 880μg/ml.We investigated the effect of flavonoids in anthriscus sylvestris on aging by using the D-galactose-induced aging mice.The result showed that high dose group reduced the content of MDA and enhance the activities of SOD in spleen and liver significantly.(4) We investigated the inhibition rate of superoxide anion free radical of polysaccharide in anthriscus sylvestris by using pyrogallol autoxidation method.The result showed that polysaccharide in anthriscus sylvestris had remarkable anti-free radical activity with the inhibition rate of 31.22%,when the concentration was lmg/ml.We evaluated the immunomodulating effects of polysaccharides from anthriscus sylvestris through carbon clearance test.Results showed that both middle and high dose groups could increase the index of K and a,the index of spleen and liver in carbon clearance test.water decoction and petroleum ether extract of anthriscus sylvestris had no obvious toxic response to the mouse under certain concentration,flavonoids in anthriscus sylvestris had remarkable in vitro and in vivo antioxidant activity when polysaccharide from anthriscus sylvestris had obvious in vitro antioxidant activity and immunomodulatory effect.Our work provides evidence for the research of the pharmacodynamic action,mechanism and exploitation of Anthriscus sylvestris.
