OBJECTIVE:Through animal experimentation we observe the water extraction (WET)and the acohol extraction(AET) of turmeric's effection to the acetic acid injurily ulcerous colitis (UC), and distinguishes its anti-UC related cell factor. From the angle of oxygen free radical (OFR) reaction we can reveal the water extraction and the acohol extraction of turmeric's mechanism in anti-UC function.METHODS:Randomly divided 180 SD rats into nine groups,20 rats every group:control group, modle group, SASP group and drug groups (WET high-dosage, middle-dosage, low-dosage group and AET high-dosage, middle-dosage, low-dosage group).SASP group:intragastric administration SASP(180mg/kg·d-1),once a day. Drug groups:intragastric administration WET (80mg/kg-d-',40mg/kg·d-1,20mg/kg·d-1)and AET(80mg/kg·d-1,40mg/kg·d-1,20mg/ kg·d-1), once a day.Every rat treated for 2 weeks, then execute by decapitation at the third week. Calculate the mucous membrane of colon's prob-morph damage fraction mark.Collect 4ml blood, detect the blood plasma malondialdehyde(MDA) contents and the blood plasma superoxide dismutase (SOD) vitality. Recipe 8cm colon architecture near the anus, conventional take HE dyeing pathological section and photographs, Calculate the mucous membrane of colon's histology damage fraction mark.Immunohistochemistry (IHC) examines Spasmolytic Polypeptide (SP) expression in colon mucous membrane.RESULTS:①The prob-morph damage and histology damage fraction marks and the blood plasma MDA contents of UC model group rats are all elevated than the normal group rats, the blood plasma SOD vitality of them droped, and the colon mucous membrane SP expressions upward.②The prob-morph damage and histology damage fraction marks and the blood plasma MDA contents of AET high-dosage and middle-dosage group is obviously lower than the model group, the blood plasma SOD vitality is obviously higher than the model group,the SP expression in colon mucous membrane is obviously down-regulated (P<0.01),and have no statistically significant differences from the SASP group also (P>0.05); The prob-morph damage and histology damage fraction marks and the blood plasma MDA contents of AET low-dosage group and WET high-dosage group is obviously lower than the model group,the blood plasma SOD vitality is obviously higher than the model group,the SP expression in colon mucous membrane is obviously down-regulated (P<0.01),but have obviously statistically significant differences from the SASP group (P<0.01) The prob-morph damage and histology damage fraction marks,the blood SOD vitality and the SP expression in colon mucous membrane of WET middle-dosage group and WET low-dosage group have no statistically significant differences from the model group (P>0.05)CONCLUSION:①Our experiment confirmed that the intestinal perfusion modeling method of ulcerative colitis lead by acetic acid injuried is reliable.②high-dosage (80mg/kg·d-1)and middle-dosage (40mg/kg·d-1) AET can promote ulcer healing in rats with ulcerative colitis lead by acetic acid injuried, the mechanism of which is enhance the SOD vitality, reduce the MDA contents, improving the body's ability to eliminate OFR, increasing the stability of colonic mucosa, thereby enhance the ability of colon mucosa in anti-OFR damage, reducing lipid peroxidation damage, prevente the generation of ulcer. The expression of SP has close relation with the ulcerous colitis mucous membrane repair, it is paralleled with the damage of colon mucous membrane.③WET can't promote ulcer healing in rats with ulcerative colitis lead by acetic acid injuried, but the large dose of which (80mg/kg·d-1) still have the function.
