The Exepresion Of Livin And SMAC Protein And The Analysis Of The Prognosis Factors In Medulloblastoma

Objective:To examine the expression of Livin and SMAC protein in medulloblastoma,discusses the carcinogenesis; and analysis the prognostic foctors of medulloblastoma, for finding a new way for medulloblastoma's treatment and judgment prognosis.Methods:The specimen about this research includes 45 samples using formalin-fixed and paraffin-embedded tissues from medulloblastoma patients after the surgery excised, and uses 10 samples of normal cerebellar tissues for comparison research. Uses the PV two step immunohistochemistry to examine the expression situation of Livin and SMAC protein in the specimen tissues, and analyzed the relationship between their expression and clinical prognosis by corresponding statistical methods.Results:Livin protein expressed 82.2%(37/45) in medulloblastoma samples, and 0%(0/10) in normal cerebellar tissues,there had significance difference betreen them (P<0.05);SMAC protein expressed 55.6%(25/45) in medulloblastoma samples, and 90%(9/10) in normal cerebellar tissues,there had significance difference betreen them so. There was prominent negative correlation between Livin and SMAC protein in medulloblastoma(R=-0.72,P<0.05)。Livin and SMAC protein's expression was not obvious differences with clinical factors on patient's sex, age, tumor spot, hydrocephalus and pathology, but it was closely related with tumor's size. Kaplan-Meien exists ananlysis and log-rank test indicated that the prognosis of the Livin positive expression group was worse than the negative expression group, while the SMAC negative expression group was worse than the positive expression group (P<0.05).COX regression analysis showed that the expression of Livin,the degree of tumor resection and postoperative treatment are the independent risk foctors of prognosis in MB patiets.Conclusions:1. Livin and SMAC protein expressed abnormally in medulloblastoma, the elevated expression of Livin and depressed expression of smac possibly cooperate each other to play an important role in medulloblastoma's occurrence and development. 2. The expression of Livin and SMAC protein were related with the patient prognosis,and they can take as the targets on the prognosis of medulloblastoma.3. Livin and SMAC are expected to become new moleculer targets in the medulloblastoma's gene therapy.4. The expression of Livin, the degree of tumor resection and postoperative treatment are the independent risk foctors of prognosis in medulloblastoma patiets.
