Objective: This study was to investigate the expressions of Livin and Smac/DIABLO and their correlations to the clinicopathological para-meters in gastric cancer.Methods: The expression levels of 2 Livin splicing variants (Livinαand Livinβ) and Smac mRNA in 42 gastric cancer specimens and 20 para-cancerous normal gastric tissues(the control) were detected by the SYBR green I real-time fluorescent quantitative reverse transcription PCR, the protein expressions and location of Livin and Smac were measured by immunohistochemistry (SP method).Results: 1. The relative mRNA expressions of Livinαand Livinβwere significantly higher in gastric cancer tissues than that in the control (P<0.01), the relative mRNA expressions of Livinαand Livinβwere no significant difference in gastric cancer tissues (P>0.05), the relative mRNA expression of Smac/DIABLO was significantly lower in gastric cancer than that in the control(P<0.01), and the protein expressions of Livin and Smac were consistent with that of their mRNA expressions. 2. The expression of Livin was higher in groups with lymph node metastasis, low differentiation and TNM stage III+IV than that in groups without lymph node metastasis, middle/high differentiation and TNM stage I+II (P<0.05); the expression of Smac was higher in TNM stage I+II group than that in TNM stage III+IV group (P<0.05). 3. The expression of Livin was negatively related to Smac in gastric cancer tissues, but correlations were not significant (r=﹣0.268).Conclusions: 1. The relative mRNA and protein expressions of Livinαand Livinβare significantly increased in gastric cancer compared with the control, the relative mRNA and protein expression of Smac is significantly decreased in gastric cancer than that in the control. 2. The expression of Livin in gastric cancer tissues has correlations to lymph node metastasis, differentiation and TNM stage, The expression of Smac in gastric cancer tissues has correlation to TNM stage.
