Objective:To observe the effects and significance of deguelin on the expression of hypoxia inducible factor-la (HIF-1α) expression in the retinas of diabetic rats, to explore the effects of the development of deguelin diabetic retinopathy.Method:1.30 SD rats were divided into groups at random using number table, completing randomization, the number of the controlling group was 10, and the remaining 20 SD rats application of streptozotocin (STZ) intraperitoneal injection of 60mg/kg established diabetic model.2.From the 1st week after the model was made successfully, deguelin intravitreal injection every a week. Right eye (the eye deguelin group) regarded as the experimental eye, left eye (diabetic eye group) regarded as the eyes of diabetic control, left eye and right eye (normal eye group) of diabetic group and normal group were injected with the same amount of empty vector.3.10 weeks later,observe the rats retinal immunohistochemically, respectively HIF-la expression and localization, electron microscopy and structural change of retinal ELISA determination of the expression of HIF-1α.Results:Immunohisochemistry showed that the HIF-1αpositive cells in retinas in diabetes eye group and deguelin intervention eye group were both higher than the normal control group,and the deguelin intervention eye group was lower than diabetes eye group.Elisa showed that the expression of HIF-1αin retinas in diabetes eye group and deguelin intervention eye group was significantly higher than those of normal control group (P<0.05),the deguelin intervention eye group was lower than diabetes eye group(P<0.05).Electron microscopy demonstrated that the pathological changes of diabetic retinopathy were observed in diabetes group and deguelin intervention eye group, the pathological degree in deguelin intervention eye group was slighter than those in diabetes eye group. Conclusion:(1) There in early diabetic rat retina the expression of HIF-la, which shows the incidence of hypoxia in the diabetic retina, may play an important role in the development.(2) After intervention deguelin reduced expression of HIF-la, indicating that rotenone always delay the process of early development of diabetic retinopathy.
