Objective:Established animal model is the indispensable research approach to explore the pathogenesis and treatment of abdominal aortic aneurysm (AAA). Yet there is rare related report of the establishment of the abdominal aortic aneurysm model in rabbit in China.This paper reports a simple and high-successed rubbit model of AAA,laid the foundation of further test the treatment of abdominal aortic aneurysm research.Methods:20 healthy male New Zealand white rabbits were divided into experimental group and control group(10 each),laparotomized after the rabbit was anesthesitized,the experimental group were underwent CaCl2 infiltration method using 0.75mol/L CaCl2 solution infiltrate the AAA 30min,then rinse and the abdomen was closed.The control group were underwent saline infiltration., using saline infiltrate the AAA 30min, then rinse and close the abdomen. Two group animals were observed expansion rate of abdominal aorta, morphological changes and pathological changes After 6 weaks.Results:The expansion rate of abdominal aortic aneurysm models exceeded 100% in the experimental group, which represented a statistically difference when compared with the control group. HE staining showed significant damage to the artery wall, inflammatory cell infiltration in the whole layer of the aortic wall;VG staining and Masson's staining showed aortic medial elastin tissue severely damaged, and poorly, losing continuity, elastin was significantly thinner, most of the broken, fragmented or absent,which conform to the histopathological changes of AAA.About the control group, no significant expansion of rabbit abdominal aorta, aneurysms do not form, and no abnormal pathology.Conclusion:The CaCl2 infiltration method to establish AAA model in rabbit is simply,easily operated and high-successed,it exhibits several histologic similarities with human abdominal aortic aneurysm,can provide a suitable animal model for the further study.
