Objective:To observe the morphological change of allogeneic rabbit abdominal aortic aneurysm(AAA) patch model and the expression changes of osteopontin(OPN)and C-reactive protein(CRP)in local arterial wall,then study the significances of them in the pathological process of abdominal aortic aneurysm(AAA).Methods:Take a total of 24 adult New Zealand white rabbits,in the experimental group below the renal arteries and above the iliac artery bifurcation plane about 1 cm,cut an incison of about 4 mm along the direction parallel to the abdominal aorta,then the body of a normal abdominal arterial wall graft patch is sutured to the recipient abdominal aorta.The allogeneic rabbit abdominal aortic aneurysm patch model is finished. The arterial wall was harvest after 7day,21days,35days respectively. The control group also blocked blood vessels in the area like the experimental group but not work patch operation.The morphological parameter of abdominal aorta was measured before operation and after operation. The level change of OPN and CRP were observed by immunohistochemical methods. Results:The rabbits in the control group and experimental groups were alive,blood flow was unobstructed in the abdominal aorta, no stenosis in stoma. The diameter of aneurysm was gradually increased with time in experimental groups,that there were significantly different (P<0.05)compared to the contronl group.The content of elastin, collagen fibers and SMC in media was in a significantly negative correlation mode with the diameter of aneurysm(P<0.05).The immunohistochemical staining showed that the expression of OPN and CRP in experiment groups were significantly higher than the control group(P<0.05).Conclusion:1 Allogeneic rabbit abdominal aortic aneurysm patch model is a reliable animal model to study the pathogenesis of abdominal aortic aneurysm.2 The expressions of OPN and CRP are increasing in the local aorta wall of allogeneic rabbit abdominal aortic aneurysm patch model.3 OPN may play a important role in the abdominal aortic aneurysm formation and development at the time of vascular remodeling and change of hemodynamics.
