Effect Of Gamma Globulin On NF-κB And MDA Of The Brain Tissue Following The Focal Cerebral Ischemia-reperfusion In Rat

Abstract/Summary:
Objective:To illustrate the neuroprotective effect of gamma globulin on cerebral ischemia-reperfusion injury in rats and to explore the possible mechanism.Methods:90 adult Wistar rats with masculinity,weighing 250-280g,were randomly divided into 3 groups:sham-operation[sham-operation combined with normal saline (NS)];experimental control group [operation combined with normal saline (NS)] and experimental group [operation combined with gamma globulin]; rat models of middle cerebral artery occulions (MCAO)-induced cerebral ischemia-reperfusion injury(MCARI) was established according to Zea Longa's method. Each group was further divided into 3 subgroups according to different phases(24h,72h,120h). After we observed the change in neurological deficit scores,we sacrificed them successively and brain tissue samples were obtained at different phases(24h,72h, 120h) in each group. We observed the range of cerebral inflarct by TTC dyeing, observed histological change in rats' brain after hematoxylin-eosin (HE) staining by the light microscope, detected the expression of NF-κB p65 in the brain injury tissue by immunohistochemistry, mensurated the content of malondialdehyde (MDA) in the brain injury tissue by the absorption spectrometry. One way-ANOVA was used for statistic analysis.Result:1. After MCARI, the experimental rats in the experimental control group and experimental group had the large range of right cerebral inflarct and neurological deficit, however, compared with the experimental control group, in the experimental group, the degree of damage was all slighter significantly at 24h,72hand 120h (P<0.05).2. The brain tissue of the experimental rats after MCARI roccurred clear pathomorphological change; however, compared with the experimental control group, in the experimental group, the degree of damage was slighter significantly at 24h,72hand 120h (P<0.05).3.The expression levels of NF-κBp65 protein in the experimental control group and experimental group were higher than those in sham-operation group at 24h,72hand 120h (P<0.05). The expression levels of NF-κBp65 protein in experimental group was lower than that of the experimental control group significantly at 24h,72hand 120h (P<0.05).4. MDA in brain tissues after MCARI raised significantly; the application of gamma globulin decreased the MDA level.Conclusion:1. The application of gamma globulin can decrease the range of cerebral inflarct and improve the neurological deficit after MCARl.2. The expression of NF-icBp65 protein and the content of MDA in the brain injury tissue increased in the acute stage of MCARI. Gamma globulin could effectively relieve the inflammatory reaction and inhibit the reaction of lipid peroxidation in the acute stage of MCARI.
