Wheat Germ Globulin is a high-quality protein,has important research value in improving physical function and eliminate inflammation.The development,utilization and extraction of wheat germ globulin are all in the field of deep processing of wheat.In this study,we studied the effects of salting-out of wheat germ globulin,in vitro biological activity analysis,and evaluation of wound tissue repair effect.This study aims to provide a theoretical basis for the systematic use of high value-added wheat germ resources,provides a guideline for the further development of functional anti-inflammatory drugs and health products and raw materials development.The main research includes the following.The effects of wheat germ globulin on the activity of mouse fibroblasts L929 were studied.The key control points in the process were studied by single factor and response surface: salt concentration,extraction temperature,The extraction time of wheat germ globulin was studied.(2)The best extraction conditions for wheat germ globulin are Salt concentration2.89%?extraction time 1.86 h?extraction temperature of 30.45 ?.Under these conditions,the proliferation index of mouse embryonic fibroblast L929 reached 67.89±2.31%.The results showed that the content of essential amino acids in wheat germ globulin was significantly higher than that in wheat germ,and the content of amino acids was balanced and the degree of dispersion was the smallest.Compared with the previous salt-soda,Osborne fractionation and salt enzyme synergistic extraction process,the bioactivity of wheat germ protein to mouse fibroblast L929 was first tested in wheat germ globulin extraction process.embryo globulin process optimization.This process greatly enhances the commercial and other application value of wheat germ globulin and broadens the application of wheat germ resource.(3)The in vitro biological activity of wheat germ globulin on RAW264.7 cells was studied.Experimental studies show that wheat germ globulin has a significant anti-inflammatory effect,a variety of concentrations of wheat germ globulin on RAW264.7cells non-toxic,can significantly promote the normal RAW264.7 NO production;The concentration of wheat germ globulin could significantly inhibit the production of NO in RAW264.7 cells induced by LPS,and the inhibitory effect was more significant with the(4)increase of concentration.Wheat germ globulin significantly inhibit pro-inflammatory cytokines TNF-?,wheat germ described globulin play a role in combating inflammation associated with inflammatory diseases proinflammatory cytokines.The proliferation of mouse fibroblast L929 was significantly increased in the range of 500 ~ 800?g / mL,and the cell proliferation index increased with the increase of concentration.From the cellular level confirmed that the wheat germ globulin in proinflammatory factors associated with inflammatory diseases play anti-inflammatory and tissue repair function.This study provides a theoretical basis for the further use of animal experiments to evaluate the effect of wheat germ protein on the repair of wound tissue.(3)The rat model of ? degree burn was established and the effect of wheat germ protein on the wound healing of mice was analyzed.Compared with the model group,the effect of wheat germ globulin on wound healing was significant(P <0.01).Histopathological observation of burn wounds showed that wheat germ globulin had obvious repair effect on wound tissue.Wheat germ globulin has significant repair of rat burn wounds,promoting wound healing;The protein content and SOD activity in the wound tissue of the model group were significantly lower than those in the normal group(P <0.01).MDA,HOP and EGF were significantly higher than those in the normal group(P <0.01),indicating that the model group had significant changes in the wound tissue;The content of protein,the content of EGF and the activity of SOD were significantly increased(P <0.01),and the content of HOP was significantly decreased(P <0.01).Experiments show that with the wheat embryo protein significantly improved the physical and chemical indicators of the wound tissue for the recovery of wound tissue provides a good condition;Through the study of immune function in burned rats,the immune dysfunction of burned rats,wheat embryo globulin significantly reduced the burns caused by the immune organs of the swelling,the immune organs have a protective effect;On the burned rat peripheral blood lymphocyte subsets have a significant regulatory role.Wheat germ globulin can significantly improve immune function in burned rats.In short,the research content of this topic for the development of functional health care products and anti-inflammatory drug raw materials to provide a theoretical reference.
