Objective:To identify the HLA-DQA1*0302,DQB1*0303 alleles associated with vitiligo in the Uygur nationality of Xinjiang region.Methods:(1) Collect the 0.5ml peripheral blood of every subject,using the DNA extract kit of of sangon biotech Co Ltd to extract peripheral blood DNA, then using 0.7% agarose gel electrophoresis check the DNA quality;(2)Polymerase chain reaction sequence-specific primer (PCR-SSP) was used to analyze the distribution of HLA-DQA1*0302,DQB1*0303 alleles among 300 patients with vitiligo and 300 healthy controls in the Uygur nationality of Xinjiang region. (3)Take the PCR production 6ul misce bene with bromchlorphenol blue 2ul, then using 1.5% agarose gel electrophoresis(contain EB). Then observe the result by the ultraviolet transmission fonnatter and gather the pictures;(4)Then check the sequence of PCR product,the results of sequence analysis by online BLAST; (5)Data statistic by the software SPSS17.0 statistics.Results:(1)The frequence of DQA1*0302 (0.2050 to 0.1383) and DQB1*0303 (0.3017 to 0.1333) alleles significantly increased in the patients with vitiligo compared with those in the controls [OR=1.817 (95%CI:1.291-2.557); OR=4.183 (95%CI:2.963-5.906) (P< 0.01)];(2)HLA-DQA1*0302 and HLA-DQB1*0303 alleles increased in both adult cases and childhood cases[childhood:OR= 1.961 (95%CI:1.221-3.148); OR=3.517 (95%CI:2.190-5.650) adult:OR= 1.750 (95%CI:1.199-2.555); OR=4.559 (95%CI:3.109-6.685) (P<0.01)];(3)The frequence of HLA-DQB1*0303 allele increased in patients with positive and negative familiy history of vitiligo[OR=1.822(95%CI:1.280-2.592) (P<0.01)]. HLA-DQA1*0302 allele increased in patients with negative familiy history of vitiligo[OR=2.903(95%CI:1.451-5.808); OR=3.744(95%CI:3.088-6.301)(P<0.01)];(4)The frequence of HLA-DQA1*0302 allele was significantly increased in the scattered and acral types of vitiligo[OR=1.795 (95%CI:1.178-2.737); OR=2.277 (95%CI:1.282-4.054) (P< 0.01)]. The frequence of HLA-DQB1*0303 allele was significantly increased in all types of vitiligo[localized:OR=3.63 (95%CI:2.034-6.479); generalized:OR=12.38 (95%CI:2.618-58.503); scattered:OR=3.775 (95%CI:2.476-5.757); acral:OR=5.645 (95%CI:3.082-10.338); segmental: OR=3.732 (95%CI:1.787-7.793) (P<0.01)].Conclusion:(1)HLA-DQA1*0302 and HLA-DQB1*0303 alleles increase the risk of suffering vitiligo,HLA-DQA1*0302, DQB1*0303 alleles may be the susceptible alleles of vitiligo in the Uygur nationality of Xinjiang region. (2)There may be different genetic backgrounds between childhood and adult patients,and between positive familiy history and negative familiy history patients,and in different types of vitiligo.
