Wikstromia indica is the dry root bark of Wikstromia indica (L.) C.A Mey., it widely distributes in the south of the Yangtze River in China and Southeast Asia and it is a folk herbal medicine clearing heat, detoxifying, diminishing swelling and pain. It is mainly used to cure bruises and traumatic injury. It can be used in bronchitis, pneumonia, mastitis and other treatment on clinical. For further research on the material basis of its efficacy and pharmacological activity, this paper did a research on its chemical constituents, analgesic and anti-inflammatory activities.First, by using ethanol, the different polarity solvents extracting, combined with active tracking methods, the two effective parts, ethyl acetate and n-butanol extractions, were studied on the chemical constituents. Using Silica column chromatography and Sephadex LH-20 column chromatography analysis repeatedly and combining with the preparative liquid chromatography, twenty three compounds were isolated, by using mass spectrometry, nuclear magnetic resonance spectroscopy and other modern techniques of photographic identification and combining with the literature, eventually twenty one compounds were identified; Secondly, using the mouse hot plate, acetic acid writhing and xylene-induced mouse ear swelling, the ethanol extraction of Wikstromia indica and the effective parts were studied about the analgesic and anti-inflammatory activities.Research on chemical constituents showed that the twenty one compounds had been identified:the five coumarins, umbdheforne(5), aphnogitin (6), daphnoretin(7), daphnoretin-7-O-β-D-glucoside(8), wikstrosin(9), respectively; the ten flavonoids, Naringin(1),5,6,7-Trihydroxy-4'-methoxy-dihydroflavonol(2), genkwanin(10), quercetin(11), quercitrin (12), sikokianin B (13), sikokianin C(14), stelleranol (15), genkwanol C (16), genkwanol B (17); the lignan compound was lirioresinol B (18); the two steroidal compounds are Daucosterol(20) andβ-sitosterol(21); the other three compounds respectively are p-hydroxybenzoic acid(3), benzoic acid(4), dibutyl phthalate(19). The compound (1) to (4) were isolated from the plant for the first time.In pharmacological studies, through the mouse hot plate test, acetic acid writhing and ear edema induced by xylene, the high, medium and low dose groups of ethanol extractions were studied on the pharmacodynamics, the results show that:(1) the ethanol extract of Wikstromia indica can prolong the pain threshold and reduce the number of writhing in mice, inhibit the inflammatory response induced by xylene and significantly reduce the ear swelling in mice, it had good analgesic and anti-inflammatory effects and showed some dose-effect relationship compared with the control group with statistical significance (P<0.05); (2) screening for the five extractions of petroleum ether, chloroform, ethyl acetate, butanol and water, the results showed that the ethyl acetate and n-butanol ones had good analgesic effect, compared with the control group with statistical significance (P<0.05), whereas petroleum ether one had some toxicity; (3) researching on 10,30,60and 90 percent ethanol elution positions of the butanol extraction further, the results showed that 60 and 90 percent of ethanol site had good efficacy compared with the control group with statistical significance (P<0.05). The results showed that the ethyl acetate and n-butanol extractions of Wikstromia indica has good analgesic effect.According to the research about the chemical constituents and pharmacological activity of Wikstromia indica, the further developments were laid a solid theoretical foundation.
