Atherosclerosis (AS), and the complication, cardio-cerebrovascular diseases, are the major causes of cardio-cerebrovascular accidents in clinical. The primary pathological basic is that the fibrous cap of vulnerable plaque (VP) rupture or fracture, and then lead to thrombus formation. So how to stabilize the atherosclerotic vulnerable plaque becomes focus in clinical and experimental research. At present, the research about the model of AS have been developed, but the ideal animal model of vulnerable plaque and/or plaque rupture is developing. This trial wants to establish a simple and practicable animal model of vulnerable plaque and evaluate the stability of plaque. After this trial, we take another to investigate the effect of formula on stabilizing the plaque, and explore initially the mechanism by which it can stabilize the plaque.The first trial:To establish and evaluate the model of atherosclerotic vulnerable plaque of carotid artery in ratsObjective:To investigate the effect of three different factors on vulnerable plaque formation of carotid artery in rats, and discuss initially intervention of drugs on stability of vulnerable plaqueMethod:With three factor, High lipid,2%L-methionine, carotid artery balloon injury, and their different combination, to establish the rat model of atherosclerotic vulnerable plaque of carotid arteryResults:In contrast to negative-control group, the level of blood lipid disorder, plasma homocysteine (Hcy), serum high-sensitivity C-reactive protein (hs-CRP) all increased with different degree, atherosclerotic vulnerable plaque of carotid artery with different degree all formed in each model group. atherosclerotic vulnerable plaque with three factors is remarkable, the concentration of hs-CRP rised (p<0.01) the plaque area is 79680.25±46517.58μm2, the ratio of stenosis is 48.02%. Each drug groups all have the effect on lessening the atherosclerotic vulnerable plaque of carotid artery.Conclusion:The concentration of serum hs-CRP in the model group due to three factors is one time more than the negative-control group, it indicates the plaque tend to rupture. The atherosclerotic vulnerable plaque of carotid artery in the model group due to three factors is obvious, it indicates there are synergistic effect among hyperlipemia, homocysteine(Hcy), and vascular endothelial dysfunction, and they work together to make the formation and progress of the atherosclerotic vulnerable plaque of carotid artery. So we choose the atherosclerotic vulnerable plaque of carotid artery due to three factors as the final model to do further research.The second trial:The invention of HUATANQUYUJIEDU formula on stability of atherosclerotic vulnerable plaque of carotid artery in ratsObjective:To investigate the effect of HUATANQUYUJIEDU formula on stability of atherosclerotic vulnerable plaque of carotid artery in rats, and explore initially the mechanism by which it can stabilize the plaque.Method:To investigate the effect of HUATANQUYUJIEDU formula on stabilizing the plaque by modeling with three-factors, and comparing with two positive-control groups.Results:The blood lipid, plasm Hcy, serum MMP-9 of the model group rised up obviously. The formation of atherosclerotic vulnerable plaque of carotid artery in the model is obvious, the ratio of stenosis is 72.60%. The expression of MMP-9 detected by immunohistochemistry is enhanced. In contrast to the model group,HUATANQUYUJIEDU formula can decreased the concentration of serum TC and LDL-C (p<0.05), decreased the concentration of plasm Hcy (p<0.01) and serum MMP-9 (p<0.01), increased the concentration of TIMP-1 (p<0.05), the expression of MMP-9 detected in the plaque is also enhanced (p<0.05), HUATANQUYUJIEDU also can reduce the ratio of MMP-9/TIMP-1 (p<0.01) Conclusion:HUATANQUYUJIEDU formula has the effect on stability of atherosclerotic vulnerable plaque in rats, it is related to mediate the blood lipid metabolism, decrease concentration of plasma Hcy, and balance the degradation of matrix in the plaque.
