Polysaccharide, as the natural high molecular compound, is a kind of Chinese traditional medicine active component. Many foreign scholars carry on researches by extracting plenty of polysaccharides from different creatures and it shows that the material has an effect on specific and non specific immune stimulation of body with a good immunocompetence. Polysaccharide works well on buildup immunity with an efficacy of anti-tumors, anti-virus, anti-bacteria and it can enhance cytotoxicity as well as promote cell factor secretion as reported in researches.In this paper, the writer uses Astragalus and Lentinan on the mice immune experiments to evaluate and compare the immune effects in different ways, such as visceral index, macrophage cell ability, delayed type hypersensitivity (DTH), NK cell activity, lymphocyte proliferation, cytokine secretion and vaccine. The writer preliminarily make sure the effective concentrations about polysaccharide and provide a rationale for using the material reasonably and screening the immunoenhancement.1 We disccussed the linear conformity degree and accuracy of polysaccharides by phenol hydrate-sulfuric determination. The results showed that the concentration was between 0.01-1.0mg/ml, a good linear relationship between the concentration of purified polysaccharides and the absorbance of reacting solution by phenol hydrate-sulfuric determination was appeared. The concentration can be relatively accurate measured when the polysaccharides without other impurities. There was an evident difference when compared to the crude extract or add protein in purified polysaccharides, suggested this method was only suitable for pure polysaccharides determination.2 To evaluate the effects on the immunity of mice with these two types of polysaccharides, we measured the visceral organs index of mice, macrophages' phagocytosis, delayed type hypersensitivity (DTH), lymphocyte proliferation capacity, NK cell activity and some Thl and Th2 cytokines. The results showed that these two types of polysaccharides can obviously enhance the delayed type hypersensitivity, strengthen macrophages' phagocytosis, increase NK cell activity, promote cytokine secretion and stimulate a higher level of antibodies for mice. These two polysaccharides also influenced the Thl/Th2 immune balance, it can significant stimulate the immune function of the mice with impaired immunity. Those two kinds of polysaccharides can raise both the Thl/Th2 immunity simultaneously and improve the Thl/Th2 cytokines secretion to prevent cyclophosphamide immunosuppression3 In addition, we found that vaccine coupled with polysaccharide can stimulate the body to produce antibodies compared to the single vaccine.To check the acute toxic side effects of polysaccharides, the mice were injected high doses of polysaccharide solution intraperitoneal for 15 days. According to the clinical research and blood biochemical parameters, the mice had no obvious symptoms when the dose was less than 0.8g/kg (equivalent to 60 times of a normal dose) and the blood biochemical parameters still stayed in the normal range, which proves that polysaccharide shows unobvious toxici side effects.
