| Objective:To determine the association between cold-heat characterization and energy metabolism in rat model of cold syndrome with the root of Aconitum Carmichaeli(Fuzi) decotion; analysing its effect trend, character and regularity. These studies explore objective marker of animal model, reveal inherent connections for cold-heat nature drugs, and establish basis for better clinical drug use.Methods:Wistar rats were given orally traditional Chinese medicine decoction (Huangqin, Huanglian, Huangbo, proportion is 1:1:1) for 14 days, then therapy with Fuzi decoction for 10 days, observing general condition, body temperature, changes in cold or heat trend etc. in 3,17,27 day respectively. Using enzyme-linked immunosorbent assay for rat plasma T3, T4, TSH, TRH content, studying liver tissue ATP enzyme activity, viscera index; using high performance liquid chromatography for liver tissue ATP and energy charge in rats.Results:The cold drugs of Huanglian, Huangbo, Huangqin taken orally can significantly extend the time in the heat box, depress plasma T3, T4, TSH levels, reduce ATP enzyme activity, and ATP content in rat liver, restrain kidney and brain growth. The drug Fuzi can elevate plasma T3, T4, TSH levels, increase ATP activity, and ATP, it is conducive to the liver, kidney brain growth.Conclusion:The growth and development of rat can be affected by continued given orally cold or heat nature drugs. It appears changes in physiological metabolism and digestive function; and different variety in behavior for cold or heat syndrome character respectively; also affecting the body endocrine and biochemical enzymes. Cold-heat nature of TCM is correlation with the trend of cold or heat properties, T3, T4, TSH, ATP and energy charge, viscera index etc.. Cold nature drugs such as Huanglian, Huangbo, Huangqin can reduce the body's energy reserves, decrease the body's energy metabolism; heat nature drug as Fuzi can increase the body's energy reserves, improve the body energy metabolism. |
