Research background and purposeAt present, the massive experiments have already confirmed that the umbilical cord blood hematopoietic stem cell has more potential of the self-renewal, the multiplication differentiation and increases in vitro than the hematopoietic stem cell which originates from the marrow and circumference blood, widely applies in the stem cell transplant as well as other clinical cares. CD34+is the marker of the hematopoietic stem cell and the most commonly used symbol of separating hematopoietic stem cell, the content of the CD34+hematopoietic stem cell is relatively high in the marrow and in the umbilical cord blood, the CD34+ hematopoietic stem cell has the strong ability of self-renewal and differentiation. And the navel blood plasma is the natural micro environment which the navel blood stem cell multiplies, in which active constituent already aroused the widespread interest. The biological chip is one kind of brand-new micro analysis technology which developed in the early 1990s, there are mainly three broad headings at present: Gene chip, protein chip, chip laboratory. Protein chip has high flux, high velocity, high grade and automation and so on, this is other examination methods can not hold a candle [1] This research use protein chip technology, analyzes the active constituent in the navel blood plasma, observes the influence to the CD34+ hematopoietic stem cell's multiplication differentiation, builds the foundation for the related cytobiology clinical practice.MethodsThis research will carries on the contrastive analysis active constituent to the navel blood plasma and the healthy youth feminine vein blood plasma by using the protein chip technology, and carries on two sample protein content difference analyses to the person navel blood plasma and the vein blood plasma with Gene Pix the Pro4.0 Software.And separates the CD34+cell from the navel blood with the MiniMACS immunity magnetism bead, and then observes the CD34+cell multiplication ability under the microscope. Statistics analysis:The statistical analysis of measurement data with SPSS 10.0 statistical software, t test, P<0.05 for the difference statistically significant.Results1 The many kinds of protein level in navel cord blood plasmas are obviously higher than the vein blood plasma. These proteins includ SCF, IL-20, IL-12, IL-3, FGF-4, BMP-2, BMP-7, IGF-1, IL-4, Frizzled-3, RAGE, CRIM-1, Neuritin, Neuropilin-2, Neurturin, SFRP-3, Tomoregulin-1 and so on;2 The navel cord blood plasma can obviously promote the CD34+cells proliferation. ConclusionThe navel cord blood plasma has many kinds of active protein ingredients, these protein level is obviously higher than the vein blood plasma; The navel cord blood plasma has the remarkable promotion action to the CD34+hematopoietic stem cell proliferation.
