Objective To investigate the distribution of CD133~+ and CD34~+ cells in human umbilical cord blood, and to provide theoretical basis for hematopoietic stem cell culture and transplantation in the treatment of malignant hematological diseases in children.Methods Collected 35 (each 20-25ml) fresh umbilical cord blood from full-term and healthy newboms after the umbilical cord cut in 30 seconds. Took 15ml placed in 20ml syringe which contained citrate glucose solution (ACD solution) and heparin 1.5ml(800u/ml). Combined the 6% hydroxyethyl(HE) starch settlement with density gradient centrifugation methods to separate the mononuclear cells. Marked the mononuclear cells with rabbit anti-mouse tag and set up the same type of double negative control IgG. At last, used flow cytometry assay technology to detect the rates of CD133~+ and CD34~+ cells of the mononuclear cells, and the rate of CD133~+ cells of CD34~+ cells, meanwhile, analyzed the correlation of birth weight and sex with them.Results(1) The rate of CD133~+ and CD34~+ cells in the mononuclear cells was 0.17%±0.08% and 0.40% ±0.16%, there was significant difference between two cells(P<0.05).(2) The rate of CD133~+ cells expressed on CD34± cells in umbilical cord blood was 78.49%± 7.53%, the rate of non CD133±cells was 19.84% ± 6.76%, and the difference was statistically significant between the two groups (P<0.05).(3) The rate of CD133~+ and CD34~+ cells was 0.18%±0.07% and 0.40%±0.17% of 19 female infants, and 0.16% ± 0.09% and 0.40% ± 0.16% of 16 male infants, and there was no significant difference between different sex (P>0.05).(4) The rate of CD133~+ cells expressed on CD34~+ cells was 78.93%±6.64% of 19 female infants and was 77.99 ± 8.66% of 16 male infants, there was no significant difference between two cells (P>0.05).(5) The mean birth weight of the 35 neonatal was 3.44 kg, and the rate of CD133~+ and CD34~+ cells cells in umbilical cord blood were not correlated with the birth weight(6) The rate of CD133~+ cells expressed on CD34~+ cells not correlated with the birth weight of newborn.Conclusion(1) The rate of CD 133~+ cells in mononuclear cells of fresh umbilical cord blood is lower than that ofCD34~+ celk(2) CD133~+ subsets cells are mainly expressed in CD34~+ cells.(3) There has no difference between the sex and birth weight of newborn in the rate of CD133~+/CD34~+ cells and the rate of CDl 33+cells expressed on CD34~+ cells respectively.
