Part I The Influence of Danshen Root on microglia activation After Spinal Cord InjuryObjective To observe danshen root and the MEK inhibitor on activation of microglia and the functional recovery of spinal cord after spinal cord injury, and to explore the contact of MEK/ERK signaling pathway and glial scar formation,or the contact of danshen root and glial scar formation.Methods Seventy-five female SD rats were randomly divided into SCI group (A group, n= 25), danshen root treated group(B group, n=25) and the U0126 treated group (C group, n=25).A, B, C group were switch to Allen's model of spinal cord injury. After injury 9 days,the C group were given intraperitoneal injection of the MEK inhibitor U0126 O.lmg/kg (with dimethyl sulfoxide (DMSO) dissolved injection), the A group were given intraperitoneal injection of the same amount of DMSO, and the C group were injected with danshen root via the abdominal cavity(lml/kg),once a day. Ten models selected in A, B, C groups were scored the nerve function by BBB scale and motion evoked potential the day before and after injury, and after 3d,7d and 28d after injury. After 1d,3d,7d and 28d,,5 rats were randomly selected in A, B, C group to obtained HE staining of spinal cord specimens, immunohistochemistry OX42(microglia marker), and to observe the activity of microglia, and calculate the number of positive cells in microscope. The data were analyzed using statistical software SPSS13.0.Results1. BBB scale The hindlimbs BBB scale of A, B and C groups 7 days after spinal cord injury was O,that was no significant difference between three groups (P> 0.05).Over time, motor function of the posterior limbs of rats showed different degrees of recovery, danshen group and U0126 group showde higher degree of recovery than the SCI group (P<0.05), and there is no significant difference between B and C groups (P> 0.05).28d after spinal cord injury the highest score of SCI group were restored to 13 points, while the U0126 was 15 points and danshen group was 16 points.2.MEP The latencies of motion evoked potential were prolonged,and amplitudes were decreased after SCI. Over time the amplitude and the latency improved in various degrees, latency and amplitude of B and C groups recover significantly (P<0.05), then there is no significant difference between B and C groups (P> 0.05).3. pathohistology After spinal cord injury, microglia activation and proliferation increased significantly, The cell activation of microglia in group A was not significantly differente, compared with C group (P> 0.05). But the cell activation of microglia in group B decreased significantly (P <0.05).Then 7d,28d after injury microglia activation of B and C groups decreased significantly compared with group A (P<0.05), while there was no significant difference between B and groups 28d after SCI(P> 0.05).Conclusion1.Danshen root can reduce the cell activation of microglia and neuronal damage after Spina Cord Injury.2. Danshen root is conducive to nerve function recovery after spinal cord injury.3. With the different of danshen root, inhibition of MEK/ERK signaling pathway could not inhibit the early microglia proliferation, but inhibit microglia proliferation later, that is conduciv to the growth of axons.4.Inhibition of MEK/ERK signaling pathway is conducive to nerve function recovery after spinal cord injury.Partâ…¡Clinical application of danshen root with posterior surgery for thoracolumbar fractures and spinal cord injuryObjective To observe the effect of danshen injection with posterior surgical treatment o thoracolumbar fractures and spinal cord injury.Methods Sixty-four patients'thoracolumbar veterbra burst fractures between May 2009 to March 2010 were Strictly selected. In accordance with the order of admission, patients were randomly divided into contral group (A group) and treatment group (B group), two groups o: patients were all treated with posterior open reduction and internal fixation of thoracolumbar fractures. After surgry, the patients of contral group were treated with routine therapy of hormore dehydration durg. The patients of treatment group were treated with danshen root injection on the basis of routine therapy.The two groups were evaluated by ASIA and JOA score before and after treatment. The data were analyzed using statistical software SPSS 13.0.Results1. ASIA classification:Compared with the preoperative situation, the treatment group and contral group were improved in one or more than one level.Then treatment group improved more significantly (p<0.05).2. JOA score:scores before and after treatment, B group was significantly better than the. group (p> 0.05).Conclusion Danshen root is conducive to recovery after the surgical treatment of thoracolumbar fractures and spinal cord injury.
