Dermatophytes. a polymorphous filamentous fungi, producing gangliospore, separated mycelium, different size of conidia and thick-walled spores, is the culprit of the infection of protein in skin and subcutaneous tissue as well as a highly-frequent pathogenic bacteria both to skin infections in human beings and animal. Chemically-synthesized medicine was widely used to cure dermatomycosis clinically, however, it has drawbacks like high price and side effects. Initial study shows that, Sophora flavescens Ait., Smilax glabra Roxb. and Dictamnus dascarpus Turcz., commonly used in traditional Chinese medicine usage, possess strong inhibition effect on candida albicans.. According to the scientific data, Cortex Pseudolaricis was added as the new antifungal agents, therefore, four kinds of Chinese herbal medicines were decided as the object of study. Under the modern extraction methods, the active ingredient was obtained. The antifungal activity and joint antibacterial activity in vitro of four herbal extracts on Microsporum gypseum, Microsporum canis, Trichophyton mentagrophytes was investigated, the optimal ratio of herbal extracts selected, and the stability of gel nature, qualitative research and content identification investigated. The main results are as follows:1. Extraction rate and antifungal activity of radix sophorae flavescentis, cortex dic tamni, rhixoma smilacis gla-brae and cortex pseudolaricis was investigated by re flux condense and M38-A broth microdilution method. The result showed four extra ction ratio is 19.54%,10.6%,14.12%and 26.05%respectively; additionally, the MI C of ethanol extract of radix sophorae flavescentis, cortex dictamni, rhixoma smilaci s glabrae and cortex pseudolaricis on Microsporum gypseum is 0.7813mg/mL,3.125 mg/mL, l00mg/mL and 3.125mg/mL respectively; on Trichophyton mentagrophytes, 0.7813mg/mL,3.125mg/mL,50mg/mL and 0.7813mg/mL respectively; on Microspo rum canis,3.125mg/mL,1.563mg/mL,25mg/mL and 0.7813mg/mL respectively; and MFC of the four ethanol extracts on Microsporum gypseum is 1.563mg/mL,6.25m g/mL,>100mg/mL and 6.25mg/mL respectively; on Trichophyton mentagrophytes, 1.563mg/mL,6.25mg/mL, l00mg/mL and 1.563mg/mL respectively; on Microsporu m canis,6.25mg/mL.3.125mg/mL,50mg/mL and 1.563mg/mL respectively. The res ults showed that the fungistasis and fungicidal effect of ethanol extracts in rhixoma smilacis glabrae is not significant, otherwise fungistasis and fungicidal effect of et hanol extracts radix sophorae flavescentis. cortex dictamni, cortex pse-udolaricis is s ignificant.2,To study the interaction among the effect of extract of traditional Chinese medicine against the common dermatophyte in vitro, and lay the foundation for the clinical usage. The checkerboard microdilution method was applied to study the effect of combining the extract of radix sophorae flavescentis, cortex dictamni,and cortex pseudolaricis against Microsporum gypseum. Microsporum canis and Trichophyton.3,Under the uniform-design method combined with pharmacodynamics, the optimal proportion of three ingredients(the ethanol extract of radix sophorae flavescentis, cortex dictamni. and rhixoma smilacis glabrae) in Transdermal preparation of Pixuanjing was selected. The uniform-design tableU9*(9+) was used and the minimal inhibitory concentration in different matching level on Microsporum gypseum was used as index. Under mathematical statistics method, the optimal propotion of the Transdermal preparation of Pixuanjing is 50mg of the extraction of radix sophorae flavescentis,90mg of the extraction of cortex dictamni and 50mg of the extraction of rhixoma smilacis glabrae.4,Composition and stability of the stroma of "pixianjing" gel was evaluated. Four factors orthogonal designed was used by selecting Carbopol-940, preparation of Chinese medicine, Glycerine and as four factors. The appearance of gel, resistance to heat and cold and accelerating centrifugal test were evaluated, and the optimal composition of the stroma was determined. The optimal preparation condition of"pixianjing"gel as follow:content of Carbopol, main drug, Glycerine and azone were 1.2%,5%,15%and 4%respectively. The"pixianjing"gel was conherent and stable, which showed no variation in heat-and-cold resistance test and no layering phenomenon under 4000r/min. the preparation techniques of pixianjing" gel was simple stable and safe.5,Establish the method of determination of content of Dictamnine in Pixianjing gel by LC02010HT. Gemini 5u C18 column with mobile phase consisting with methanol-water (55:45v/v) was adopted, flow rate was l.0mL/min, the detecting wavelength was set at 235nm, the column temperature was 30℃, the sample size was 10μL. The calibration curve showed good linearity over the range of 0.216-86.4μg (r=0.9993) with RSD 0.26%. the average recovery was 99.69%with RSD 0.57%. the method was sensitive, rapid accurate and reliable.
