| As the incidence of breast cancer rising rapidly, this malignant tumor has become a serious threat to women's lives and health in China. Although surgery, radiotherapy, chemotherapy and endocrine therapy has made great progress, but for advanced and metastatic breast cancer there is no effective treatment.With the development of molecular biology, cell biology and immunology progress,biotherapy becomes a new way for cancer treatment. Tumor cells can evade the immune system surveillance and grow split and proliferate. In human anti-tumor immune mechanisms, T cell-mediated immunity is the principal way. T cell activation requires two signal systems. Antigen-presenting cell surface MHC-peptide complex combine to the corresponding antigen-specific T cell receptor (TCR) produce the first signal. The second signal is non- antigen and passed by B7/CD28 costimulatory molecules family. In addition to generating and maintaining T cell responses,co-stimulatory pathway can also pass inhibitory signals, down or terminate T cell responses. B7-H3 is one of the B7/CD28 family and plays an important role in the anti-tumor immunity As is known,B7-H3 plays an important role in anti-tumor immunity.But previous datas show that it has both costimulatory and inhibitory functions.This paper studied the expression of B7-H3 in breast cancer and the relationship between the cancer occurrence and B7-H3 expression, in order to provide a reference for further study of anti-tumor immunity and biological therapy of breast tumor.Objective:Detection the expression of soluble ILT3 in breast cancer serum,and analysis the relationship between B7-H3 expression and the clinical parameters. Methods:Appling methods of ELISA to detect the level of soluble B7-H3 expression in 80 breast cancer serum and 30 normal serum, and to analyze the relationship between B7-H3 expression and clinic pathological parameters such as age, tumor size, histological grade, lymph node metastasis, estrogen and progesterone receptor status, HER-2 expression and so on.Results:1.The content of soluble B7-H3 in the serum of breast cancer specimens was 24.02±16.99ng/ml,significantly higher than normal specimens14.06±3.45ng/ml.There was significant statistical difference (P<0.05.2.The content of soluble B7-H3 in the serum of preoperative breast cancer was 24.02±16.99ng/ml, significantly higher than after the operation 14.78±3.19 ng/ml. There was significant statistical difference (P<0.05)3.B7-H3 high expression was related to clinic pathological parameters such as histological grade, lymph node metastasis, and HER-2 expression.Conclusion:The levels of soluble B7-H3 in breast cancer serum was significantly higher than normal specimens,and its levels was positively correlated with histological grade, lymph node metastasis, and HER-2 expression.sB7-H3 may affect tumor prognosis and progression.sB7-H3 high expression in breast tumors may mediate an important mechanism of the tumor immune escape. Post-operative serum levels of soluble B7-H3 in breast cancer decreased significantly, suggesting that B7-H3 can be used as a detection index for the diagnosis of malignant tumors and efficacy. |
