Part 1:Expression of PDGF-B in non-small cell lung cancer and relationship with microvessel densityObjective:To investigate the expressions of platelet-derived growth factor B(PDGF-B) and microvessel density(MVD) with the angiogenesis and the biological behaviour characteristics in non-small cell lung cancer (NSCLC)Methods:65 cases of non-small cell lung cancer(NSCLC) and 18 normal lung tissue with clinical data and follow-up results were collected from the patients who received operation in Jinan Central Hospital Affiliated to Shandong University between July 2008 and February 2010. Immunohistochemical method was used to detect PDGF-B and MVD expression. The relationship between PDGF-B and MVD expression with clinicopathologic features was analyzed.Results:The positive expression of PDGF-B(58.5%),MVD(43.13±15.40) in in non-small cell lung cancer were significanly higher than those in normal lung tissues(P<0.05). The positive of PDGF-B and MVD expression in NSCLC was significantly associated with tumor size, differentiation degree, lymph node metastasis and clinical stage(P<0.05 or P<0.01),but not with age,gender,smoking, pathology type and primary position (P>0.05). There was a positive correlation between expression of PDGF-B and MVD(r=0.488, P<0.05).Conclusion:The PDGF-B and MVD were higher expressed in patients with non-small cell lung cancer.A higher exprssion of PDGF-B is an positive angiogenis factor and playes an important role in the development and metastasis of non-small cell lung cancer.Part2:Expression of platelet-derived growth factor B mRNA and protein and the relationship with microvessel density in non-small cell lung cancerObjective:To investigate the expresion of platelet-derived growth factor B (PDGF-B)mRNA and protein and to evaluate its clinical significance with the microvessel density(MVD) in non-small cell lung cancer.Methods:65 cases of non-small cell lung cancer(NSCLC) and 18 normal lung tissue were collected from the patients who received operation in Jinan Central Hospital Affiliated to Shandong University between July 2008 and February 2010. Semi-quantitative RT-PCR assay and Western-blotting.were used to detect PDGF-B mRNA and PBGF-B protein expression respectively. The relationship between PDGF-B expression with clinical parameters, such as gender,length,smoking, pathology type, pathology grade, clinical stage, lymphatic metastasis,was analyzed.Results: PDGF-B mRNA and PDGF-B protein were both expressed in non-small cell lung cancer tissues and normal lung tissues. There was a significant overexpression of PDGF-B m RNA and protein in non-small cell lung cancer tissues as compared with normal lung tissues. The expression of PDGF-B mRNA and protein in non-small cell lung cancer were significanly higher than that in normal tissues(P<0.01). There was a positive correlation between expression of PDGF-B mRNA,protein and MVD(r=0.637, P<0.01; r=0.652, P<0.05). Expression of PDGF mRNA and protein were correlated with the, tumor size, pathology grades, clinical stage and lymphatic metastasis (P<0.05 or P<0.01),but not with patient's ages, genders, smoking, pathology types (P>0.05)Conclusion:PDGF-B m RNA and protein are highly expressed in non-small cell lung cancer tissues,and PDGF-B is an positive angiogenis factor and playes an important role in the development and metastasis of lung cancer.It can be consided an objective and effective maker to evaluate the level of malignancy of non-small cell lung cancer.
