objective To explore the inhibitory action on expression of lectin-like oxidized low-density lipoprotein receptor-1 and hsCRP and the antiatherogenic role of polyinosinic acid in atherosclerotic rabbits.methods Thirty male New Zealand white rabbits were randomly divided into five groups:control group, hypercholesterol group, poly I group, fluvastatin group and association group (n=6).Except control group, the others were strained by balloon on abdominal aortic after they received general anesthesia by 3% pentobarbital sodium at the beginning. Then they were fed respectively with polyinosinic acid (polyⅠ) and fluvastatin as well as hypercholesterol.After 12 weeks, rabbits were sacrificed and abdominal aortas were removed to examined pathological changes and the expressions of LOX-1 mRNA and protein in vascular tissue by immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR) respectively, hsCRP was also examined by Elisa.results Pathological changes of abdominal aortic:①control group, ascular intima endothelial cells were integrate and had no obvious lipid deposition;②hypercholesterol group, vascular intima endothelial cells fell off and there was visible atheromatous plaque formation. Under the fibrous cap, large numbers of amorphous necrotic disintegrating products could be saw with large amounts of cholesterol clefts, foam cells and lymphocytes;③fluvastatin group, endothelial cells part fell off. Beneath the vascular intima, there were scattered and irregular plaques with a small amount of foam cells;④poly I group, endothelial cells part fell off and there had a lot of foam cells below fibrous cap,as well as inflammatory cells;⑤association group, there had a obvious weaker atherosclerosis than hypercholesterol group. Its ascular intima slightly thickened, and had a little foam cells, smooth muscle ranked tidily. There were high level expressions of LOX-1 protein and mRNA in the vascular tissues of hypercholesterol group, but rarely in the control group. The expression levels of LOX-1 protein and mRNA in both fluvastatin group and association group were significantly lower than that in hypercholesterol group (P<0.01). The diffrence of expression levels of LOX-1 protein and mRNA between hypercholesterol group and polyⅠgroup had no statistical significance (P>0.05).Conclusion PolyⅠmaybe have a role to resist atherosclerosis and expresion of hsCRP, but it had not a obviously effect on reducing the expressions of LOX-1 protein and mRNA. Fluvastatin can significantly inhibit the development of atherosclerosis and the expressions of LOX-1 protein and mRNA.
