ObjectiveThe early diagnosis of invasive pulmonary aspergillosis was still difficult and antifungal efficacy was poor, so invasive pulmonary aspergillosis became one important cause of death in patients with hematologic malignancies and hematopoietic stem cell transplant recipients. This study was designed to establish the rat model of invasive pulmonary aspergillosis and assess the therapeutic effect of compound Fuzhengkemei on invasive pulmonary aspergillosis in rats.MethodsThe rats were immunosuppressed by intraperitoneal injection of cyclophosphamide, and then inoculated into trachea with a suspension of aspergillosis fumigatus spores. The rats were fed intragastrically with compound Fuzhengkemei. Galactomannan(GM) detection of blood samples and lung histopathology were performed to assess the therapeutic effect of compound Fuzhengkemei on invasive pulmonary aspergillosis in rats.Results1.After fungal inoculation,rats became poor spirit with diminished appetite,lack of movement,weight loss and so on.GM levels of blood samples on the third day after inoculation were 1.214±0.626 in rats with fungal inoculation compared with 0.360±0.106 in rats without fungal inoculation(P<0.001).2.Hyphae of Aspergillus fumigatus were identified in the lung tissue of all rats with fungal inoculation.3.GM values of blood samples after treatment in treated rats were 0.720±0.291 compared with 1.383±0.592 in untreated rats(P<0.001).4.The survival days after inoculation in treated rats were 12.9±4.8 compared with 9.9±3.2 in untreated rats (P=0.027).ConclusionThe rat model of IPA was successfully established based on the results of clinical manifestations, GM values of blood samples on the third day after fungal inoculation and hyphae of Aspergillus fumigatus identified in the lung tissue of all rats with fungal inoculation. In the research of treatment with Compound Fuzhengkemei, we found that Compound Fuzhengkemei significantly reduced GM values and prolonged the survival days of rats with invasive pulmonary aspergillosis. The improved morphology and decreased inflammation of lung tissue in treated rats were consistent with the results of reduced GM values and prolonged survival days. This study suggests that Compound Fuzhengkemei has therapeutic effect on invasive pulmonary aspergillosis in rats.
