[Objective]:To investigate the lymphangiogenesis factor [lymphatic endothelial hyaluronan receptor-1 (LYVE-1) and homeobox gene transcription factor-1 (Prox-1)] the quantitative expression and cervical lymph node metastasis relevance.[Method]:Select from September 2007 to June 2008 Shandong Tumor Hospital gynecological surgery fresh tissue specimens of cervical squamous cell carcinoma and adjacent normal tissues 24 cases, none of the line chemotherapy before surgery, postoperative pathology confirmed, and a complete clinical and pathological information. Patients aged 35 to 70 years, median age 42 years,5 cases of postmenopausal women, premenopausal 19 cases. According to the International League of Gynecology and Obstetrics (FIGO,2009) of diagnostic criteria for classification of cervical cancer,24 cases 3 casesⅠA,ⅠB 12 cases,ⅡA of 9 cases. Histologic grade:G1 5 cases, G2 6 cases, G3 13 cases; 16 patients with lymph node metastasis, no lymph node metastasis in 8 cases; there were 17 cases of lymphatic invasion, no lymph vessel invasion in 7 cases, tissue removal half-hour under the conditions of the specimens within the sterile saline wash the blood and remove necrotic part has been placed in -80℃refrigerator kept for inspection. RT-PCR technique using LYVE-1 and Prox-1 molecule expression quantitative analysis, and compare these two factors in the surgical specimens of tumor tissue, adjacent normal tissue differences.[Results]:Prox-1 in tumor and adjacent normal tissue were:1.52±0.26 and 0.46±0.19 (P<0.01), LYVE-1 in tumor and adjacent normal tissues were:1.37±0.21 and 0.56±0.12 (P<0.01), statistically significant difference. Prox-1 and LYVE-1 expression in tumor tissue than normal tissue, and tumor lymph node metastasis. Analysis of cervical cancer in LYVE-1, Prox-1 expression and clinical pathological factors showed:Prox-1 expression before and after menopause, the positive rate of no significant difference (Fisher exact test P=1.0000); Spearman rank correlation analysis Results (rs=0.0459, P=0.8314) and no statistical significance, indicating that menopause Prox-1 were correlated with menstrual status has nothing to do; Lyve-1 expression was no significant difference in positive rate (Fisher exact test P= 1.0000); Spearman rank correlation analysis (rs=0.0592, P=0.7834) and no statistical significance, indicating that menopause Lyve-1 were correlated with menstrual status has nothing to do. During the different clinical expression of Prox-1 positive rate was statistically significant difference (Fisher exact test P=0.0020), shows the different clinical stages of the expression of Prox-1 positive rate of different; Spearman rank correlation analysis revealed that Prox-1 in expression and clinical stage and positive relationship between the statistically significant (rs=0.6147, P= 0.0014), according to the results that the higher the clinical stage, expression of Prox-1 positive rate is higher; different clinical Lyve-1 expression during the positive rate was statistically significant difference (Fisher exact test P=0.0161), clinical stage shows different Lyve-1 expression rate of the different; Spearman rank correlation analysis showed positive Lyve-1 expression There was positive correlation between clinical stage and there was significant relationship (rs=0.46, P=0.0237), according to the results that the higher the clinical stage Lyve-1 positive expression rate. Between different histological expression of Prox-1 positive rate was statistically significant difference (Fisher exact test P=0.0433), histological grade shows different expression of Prox-1 positive rate of different; Spearman rank correlation analysis revealed that Prox-1 were correlated with histological grade and positive relationship between the statistically significant (rs=0.4822, P=0.0170), according to the results that the higher the histological grade expression of Prox-1 positive rate is higher; different organizations histological grading Lyve-1 expression between the positive rate was statistically significant difference (Fisher exact test P=0.0099), histological grade shows different Lyve-1 expression rate of the different; Spearman rank correlation analysis showed positive Lyve-1 expression with histological grade and positive relationship between the statistically significant (rs=0.5611, P=0.0043), according to the results that the higher the histological grade Lyve-1 positive expression rate. Lymph node metastasis of Prox-1 expression was statistically significant difference in positive rate (Fisher exact test P=0.0066), lymph node metastasis group that Prox-1 expression rate of the different; Spearman rank correlation analysis showed Prox-1 positive expression and lymph node metastasis and positive relationship between the statistically significant (rs=0.6325, P=0.0009), according to the result that the incidence of lymph node metastasis the expression of Prox-1 positive rate; lymph node metastasis group Lyve-1 expression was significantly different positive rates (Fisher exact test P=0.0069), lymph node metastasis group that Lyve-1 expression rate of the different; Spearman rank correlation analysis showed Lyve-1's expression and lymph node metastasis and positive relationship between the statistically significant (rs=0.6124, P=0.0015), according to the result that the incidence of lymph node metastasis Lyve-1 positive expression rate. Group without lymphatic invasion expression of Prox-1 positive rate was statistically significant difference (Fisher exact test P=0.0033), lymphatic invasion that group whether Prox-1 expression rate of the different; Spearman rank correlation analysis Show Prox-1 were correlated with lymphatic invasion and positive relationship between the statistically significant (rs=0.6969, P=0.0002), according to the results that occur lymphatic invasion by the expression of Prox-1 positive rate; without lymphatic invasion group Lyve-1 expression was significantly different positive rates (Fisher exact test P=0.0381), that group without lymphatic invasion Lyve-1 expression rate of the different; Spearman rank correlation Lyve-1 showed positive expression with lymphatic vessel invasion and positive relationship between the statistically significant (rs=0.4763, P=0.0186), according to the results that occur lymphatic invasion were Lyve-1 expression rate high.[Conclusion]:cervical cancer tissues Prox-1 and LYVE-1 expression with high lymph node metastasis of cervical cancer, the detection of the indicator for understanding the determination of cervical cancer invasion and metastasis and prognosis of a certain value.
