Abstract/Summary:
Part one Establishment of rat renal transplantation modelsObjective:To establish a stable, and reliable rat renal transplantation models under simple experimental equipment.Methods:Wistar and SD rats were used as the donors and the recipients respectively. In donation:The donors'left ureter, bladder,left kidney and vasculars connected to graft, such as aorta abdominalis and inferior vena cave were isolated, then grafts were flushed with 4℃lactated Ringer solution in situ. In implantation the donor inferior vena was anastomosed end-to-end with the recipient left renal vein, donor's aorta was anastomosed to recipient's aorta with end-to-side suture. The donor ureter with ureterocystic patch was sutured to the recipient's bladder.Results:The study was divided into two stages. In the pre-experiment stage, fifty cases of rat kidney transplantation were performed, animals were used for training harvesting grafts, vascular anastomoses, until a stable and liable renal transplantation model was established. In the formal experiment stage, forty rat kidney transplantation were performed, the time for donors:45±10min, for recipients:55±15min, for venous anastomosis:7±4min, for artery anastomosis: 13±6min, for the blocking of aortic abdomina:13.5±6min, for urinary tract reconstruction:5±3min, the warm ischemia time and the cold ischemia were 30±15s and 50±10min respectively. The success rate in the later experimental stage was 70%.Conclusion:Modification of venous anastomosis from donor's IVC end-to-side to recipient's IVC to donor's IVC end-to-end to recipient's left renal vein, resulted in improvement of success rat of this model, which can be preformed under simple experimental equipment. Part Two Effect of emodin on the acute rejection in rat renal transplantationObjective:To investigate the effects of emodin on acute allo-rejection in rat renal transplantation models.Methods:Wistar and SD rats were used as the donors and the recipients respectively. Recipients were divided into four groups at random, which were treated with saline, emodin, ciclosporin A, emodin+ciclosporin A from day 0 to day 7 posttransplantatively. General conditions of rats were observed after operation. Rats in each group were sacrificed at 7 days after transplantation, the pathology of grafts and levels IL-2 in serum were observed.Results:General conditions of recipients in emodin group and CsA+emodin group were better than those of control group and CsA group respectively. Serum IL-2 levels in emodin group were significantly lower than in control group. To some extend, emodin could decrease the production of IL-2 in rat allo-renal transplantation, production of IL-2 was suppressed to a lower levels in CsA+emodin group as compared with that in CsA group, the difference was significant in statistics. Same results were found in pathological examination, emodin could not suppress acute rejection in rat allo-renal transplantation. But combination of CsA and emodin could improve the effects of CsA in suppression of allo-renal rejection in rat and decrease its side effect.Conclusion:Emodin could decrease the production of IL-2 in rat allo-renal transplantation, which was not enough to suppress allo-graft rejection. Combination of CsA and emodin resulted in improvement of CsA effects on suppression allo-graft rejection and probably decreasing the side-effect of CsA.
