Objective:To observe the expressions of VIP (vasoactive intestinal peptide) proteins in the tissue of gastric carcinoma and normal gastric beside carcinoma, and NKG2D (natural killer group 2,member D),DAP10, PI3K (phosphatidylinositol 3 kinase) and NF-κB (nuclear factor-kappa B) proteins in the inflammatory cells, and to evaluate the possible relations between them.Methods:72 patients enrolled into our study,which come from the First Affiliated Hospital of Nanchang University from the September 2008 to June 2009, received gastric cancer surgery, and diagnosed by gastric tissue pathology. Collected the gastric carcinoma, normal tissue beside carcinoma and clinical patho-parameters of these patients.There were 52 men and 20 women, the age range from 40 to 83 years old, the average age is 60.4±11.4 years old. Among them,there were 34 cases range from the age of 40-60 years old, and 38 cases are grater than 60 years old; 44 cases of adenocarcinom located at the pyloric antrum and 28 cases at the body of the stomach; 44 cases had lymph node metastasis,28 cases had no lymph node metastasis; 24 cases with well-moderately differentiated,48 cases with poorly differentiated. Acorrding to the TNM,30 cases were staged in the period of TNMⅠtoⅡand 42 cases in the of period TNM III to VI. Observe the expressions of VIP proteins in the tissue of gastric carcinoma and normal gastric tissue beside carcinoma, and NKG2D,DAP10,PI3K and NF-κB proteins in the inflammatory cells.Results:l.The positive expression rates of VIP in gastric carcinoma, normal tissue beside carcinoma were 96%and 89%respectively. The former is higher than the latter, but with no significant difference between them(P>0.05).The expression intensity of VIP in gastric carcinoma was higher than the normal tissue beside carcinoma(P<0.05).The VIP expression intensity in the patients with poorly differentiated degree,having lymph node metastasis,or staged in the period of TNMⅢtoⅣ,was significantly higher than the patients with well-moderately differentiated degree, having no lymphnode metastasis, or staging in the period of TNM I toⅡrespectively(P<0.05). However,the VIP expression intensity had no significant difference in different sex, different age and different cancer region (P>0.05).2. The positive expression rates of NKG2D in the inflammatory cells of gastric carcinoma and normal tissue beside carcinoma were 44%and 96%respectively.The former is lower than the latter with significant difference between them(P<0.05).The expression intensity of NKG2D in the inflammatory cells of gastric carcinoma was lower than normal tissue beside carcinoma (P<0.01).The NKG2D expression intensity in the inflammatory cells of gastric carcinoma in the patients with poorly differentiated degree,having lymph node metastasis,or staged in the period of TNMⅢto IV,was significant lower than the patients with well-moderately differentiated degree, having no lymphnode metastasis,or staging in the period of TNMⅠtoⅡrespectively (P<0.05). However,the NKG2D expression intensity had no significant difference in different sex, different age and different cancer region (P>0.05).3.The positive expression rates of DAP10 in the inflammatory cells gastric of carcinoma and normal tissue beside carcinoma were 50%and 92%respectively. The former is lower than the latter with significant difference between them(P<0.01).The expression intensity of DAP 10 in the inflammatory cells of gastric carcinoma was lower than the normal tissue beside carcinoma (P<0.01).The DAP10 expression intensity in the inflammatory cells of gastric carcinoma in the patients with poorly differentiated degree,having lymph node metastasis,or staged in the period of TNMⅢtoⅣ, was significant lower than the patients with well-moderately differentiated degree, having no lymphnode metastasis,or staging in the period of TNMⅠtoⅡrespectively(P<0.05). However,the DAP10 expression intensity had no significant difference in different sex, different age and different cancer region(P>0.05).4.The positive expression rates of PI3K p85ain the inflammatory cells of gastric carcinoma and normal tissue beside carcinoma were 31%and 71%respectively. The former is lower than the latter with significant difference between them(P<0.01).The expression intensity of PI3K p85ain the inflammatory cells of gastric carcinoma was lower than the normal tissue beside carcinoma (P<0.01).The PI3K p85aexpression intensity in the inflammatory cells of gastric carcinoma in the patients with poorly differentiated degree, having lymph node metastasis,or staged in the period of TNMⅢtoⅣ,was significant lower than the patients with well-moderately differentiated degree, having no lymphnode metastasis,or staging in the period of TNMⅠtoⅡrespectively(P<0.05). However,the PI3K p85aexpression intensity had no significant difference in different sex, different age and different cancer region (P>0.05).5.The positive expression rates of NF-κB p65 in the inflammatory cells of gastric carcinoma, normal tissue beside carcinoma were 57%and 86%respectively. The former is lower than the latter with significant difference between them (P<0.05).The expression intensity of NF-κB p65 in the inflammatory cells of gastric carcinoma was lower than the normal tissue beside carcinoma (P<0.05).The NF-κB p65 expression intensity in the inflammatory cells of gastric carcinoma in the patients with poorly differentiated degree,having lymph node metastasis,or staged in the period of TNMⅢtoⅣ,was significant lower than the patients with well-moderately differentiated degree, having no lymphnode metastasis,or staging in the period of TNMⅠtoⅡrespectively(P<0.05). However,the NF-κB p65 expression intensity had no significant difference in different sex, different age and different cancer region (P>0.05).6.There were negative correlation between the expression of VIP in carcinoma and the expression of NKG2D,DAP10,PI3K p85a,NF-KB p65 in the inflammatory cells of gastric carcinoma (P<0.05).Conclusions:The expression intensity of VIP in gastric carcinoma was higher than the normal tissue beside carcinoma.However, The expression intensity of NKG2D, DAP10,PI3K, NF-κB in the inflammatory cells of gastric carcinoma was lower than the normal tissue beside carcinoma. There were negative correlation between the expression of VIP in carcinoma and the expression of NKG2D,DAP10,PI3K,NF-κB in the inflammatory cells of gastric carcinoma (P<0.05). During the process of occurrence and development of gastric cancer, VIP may inhibit the NKG2D/DAP10/PI3K7NF-κB pathway to participate in the immune evasion of gastric carcinoma,and promot the proliferation,differentiation,and metastasis of gastric cancer.
