Objective:The purpose of this study is to examine the expression of MCM2 and Ki-67 protein in human Endometrioid Adenocarcinoma and atypical hyperplasia of endometrium (EAH),so as to explore the role of MCM2 and Ki-67 in the tumorigenesis and progression of human Endometrioid Adenocarcinoma and speculated the existence of a mechanism in the regulation of cell proliferation.Methods:The expression of MCM2 and Ki-67 in 20 cases of normal endometrium tissue, 20 cases of atypical hyperplasia of endometrium (EAH) and 50 cases of Endometrioid Adenocarcinoma tissues were detected by immunohistochemical EliVision two-stages method.And then analyzed statistically.Results:1. MCM2 protein was detected in normal endometrium tissue,atypical hyperplasia of endometrium tissue and Endometrioid Adenocarcinoma tissues. The positive expression rate of MCM2 protein is 15%,45%and 74%respectively.The rate in atypical hyperplasia of endometrium tissue was considerably higher than that in normal tissuse (P= 0.013, P<0.05). The positive expression rate for MCM? protein in human Endometrioid Adenocarcinoma tissues was higher than that in atypical hyperplasia of endometrium tissue (P=0.021, P<0.05).the expression of MCM2was related to the histopathologic grade,depth of stromal invasion.2. Ki-67 protein was detected in normal endometrium tissue,atypical hyperplasia of endometrium tissue and Endometrioid Adenocarcinoma tissues. The rate of Ki-67 protein expression in atypical hyperplasia of endometrium tissue was considerably higher than that in normal tissuse (P=0.027,P<0.05). The positive expression rate for Ki-67 protein in human Endometrioid Adenocarcinoma tissues was not so much higher than that in atypical hyperplasia of endometrium tissue (P=0.350, P>0.05). The expression of Ki-67 was related to the lymph node metastasis, independent of other clinicopathological features, histological grade, tumor invasion.3. There was a positive correlation between MCM2 and Ki-67 protein in Endometrioid Adenocarcinoma (r= 0.368, P= 0.012)4. High expression of MCM2 notice Poor prognosis.The 3-years survival rate in low-expression group was higher than the high expression group (P=0.033,P<0.05).Conclusions:The expression MCM2 and Ki-67 protein are increasing. Their abnormal expression maybe associated with malignant biologial behavior. MCM2 and Ki-67 expression in Endometrioid Adenocarcinoma was positively correlated with each other, there may be interactions between the two. Furthermore,the expression of MCM2 may provide useful prognosis implication.
