Objective:Discussion and research the inhibition and mechanism of XIAO HUANG MO polysaccharide on H22 tumor-burdened mice transplant tumor.Method:Take XIAO HUANG MO according to alcohol precipitation method extracted polysaccharide.1. XIAO HUANG MO polysaccharides antioxidant experiment in vitro:get health kunming mice killed,take liver, made liver homogenate, detection the effect of different concentrations of XIAO HUANG MO polysaccharide on H2O2 induced curled the liver damage organization superoxide dismutase(SOD) activity,catalane(CAT)activity,glutathione peroxidas(GSH-PX) activity,malondialdehyde (MDA) content,the clearance of hydroxyl radicals and DPPH.2. XIAO HUANG MO polysaccharides in vivo inhibiting tumor experiment: take 60 health kunming mice, according to the proportion of male and female 1 to 1 random into six groups, each group of 10.Namely the normal group,model group,cyclophosphamide (CTX) group,XIAO HUANG MO polysaccharides high dose group,medium dose group and low dose group.In addition to the normal group, other group all establishing H22 tumor burdened mice transplant tumor model, and inoculation after 24h began to medicine, normal group and model group day 1 times irrigation clothing 0.5ml saline, CTX group day 1 times intra-abdominal injection 20mg/kg CTX, XIAO HUANG MO polysaccharides high dose group,medium dose group and low dose group day 1 times 200mg/kg, 100mg/kg,50mg/kg irrigation for medicine stomach,continuous medication 11 days and each weight change detailed records in mice.The 12 days after picking eyeball take blood neck dislocated killed the mice, take tumor,liver,splenic organ and thymus gland, calculation inhibiting tumor proportion,spleen index,thymus index; Detection liver superoxide dismutase (SOD) activity, catalane (CAT) activity, malondialdehyde (MDA) content, glutathione peroxidas (GSH-Px) activity,glutathione(GSH)activity; Determination of cytokines interleukin-2 (IL-2) level,tumor necrosis factor-α(TNF-α) level,vascular endothelial generation factor (VEGF) level; Determination blood Serum albumin (ALB) content; Calculation white blood cell (WBC) amount. Results:1. XIAO HUANG MO polysaccharide antioxidant experiment in vitro suggesting that its has certain oxidation resistance, and the higher the concentration, its antioxidant ability stronger.2. XIAO HUANG MO polysaccharides in vivo inhibiting tumor experiment:(1) Compared with CTX group, XIAO HUANG MO polysaccharide each dose group mice were significantly increase the weight Compared with model group, XIAO HUANG MO polysaccharide each dose group and CTX group tumors in mice were significantly reduce weight. XIAO HUANG MO polysaccharide medium dose group and CTX group inhibiting tumor the highest rate be 39.13%, high dose group and low dose group be 30.43%,34.78%; (2)Compared with CTX group, XIAO HUANG MO polysaccharide each dose group all the spleen index can improve the mice; (3) Compared with CTX group and model group, XIAO HUANG MO polysaccharide high dose group and low dose group are able to different degree increase mice liver tissue SOD,CAT,GSH-PX,GSH activity. XIAO HUANG MO polysaccharide each dose group mouse liver tissue were able to reduce MDA content:(4) Compared with model group, XIAO HUANG MO polysaccharide high dose group and medium dose group mice increased serum albumin content Compared with CTX group, XIAO HUANG MO polysaccharide each dose group mice all can increase the amount of WBC; (5) Compared with CTX group, XIAO HUANG MO polysaccharide each dose group mice increased serum IL-2 levels, reduce VEGF level. Compared with model group, XIAO HUANG MO polysaccharide high dose group mice have elevated serum TNF-a level.Conclusion:XIAO HUANG MO polysaccharide to H22 tumor burdened mice transplant tumor has certain inhibition, its action mechanisms may instead of can enhance immunity and antioxidant related.
