Objective:To study the infection of human herpes virus 8 (HHV-8) in squamous cell carcinoma, and then to clear the distribution of Human herpesvirus 8 ORF26 and ORF75 subtypes in squamous cell carcinoma.Methods:Paraffin-embedded tissue specimens of 40 cases of skin squamous cell carcinoma and 46 of esophageal squamous cell carcinoma were extracted HHV-8 DNA, amplification, direct sequencing, using DNASTAR software, Crustal W and PHYLIP software packages on the sequencing results of phylogenetic analys. To determine the HHV-8ORF26, ORF75 isoform, and finally usingχ2 test and Fisher exact test results were statistically analyzed.Methods:ORF26 test results:①40 cases of skin squamous cell carcinoma,there are 9 cases were positive for HHV-8 infection, the positive rate was 22.5%;46 cases esophageal squamous cell carcinoma,10 cases were infected by HHV-8, the positive rate was 21.7%.②The subtype HHV-8 ORF26 of skin squamous cell carcinoma 7 were subtype A, subtype C in 2 cases; esophageal squamous cell carcinoma subtype infection of HHV-8 ORF267 were subtype A,3 were C subtype,③infected with HHV-8 ORF26 squamous cell carcinoma subtypes and squamous cell carcinoma of the site of infection and pathological grade was not statistically significant (P> 0.05); (ORF75 test results)①40 cases of skin squamous cell carcinoma,10 cases of HHV-8 infection is positive, the positive rate was 25.0%; 46 cases,12 cases of esophageal squamous cell carcinoma, the positive rate was 26.1%.②skin squamous cell carcinoma subtype of HHV-8 ORF75 infection in 6 cases for the A subtype,4 cases of subtype C; esophageal squamous cell carcinoma subtype infection of HHV-8 ORF75 8 cases of subtype A,4 cases of squamous cell carcinoma③subtype C infection of HHV-8 ORF75 is forms in patients with squamous cell carcinoma and the pathological grade was the site of infection was not statistically significance (P> 0.05).Results:Squamous cell carcinoma patients infected with HHV-8 ORF26, ORF75, and C subtypes are subtype A subtype; squamous cell infected with HHV-8 ORF26, ORF75 subtypes and squamous cell carcinoma of the site of infection and pathological grading.
