Objective:To investigate the effect of ardipusilloside I on the proliferation and apoptosis of mucoepidermoid carcinoma Mc3 cells and explore its anti-tumor mechanism.Methods:The inhibitory effect of ardipusilloside I of Mc3 cells was measured by MTT assay. The apoptotic ratio and cell cycle analysis were determined by flow cytometry. Cell apoptosis was observed by fluorescence microscope techniques with Hoechst 33342 associated with TEM. DNA ladder was observed through agarose gel electrophoresis. Western blotting was used to assay changes of the proteins correlated cell apoptosis such as Bcl-2 /Bax and caspase-3.Results:ADS I significantly inhibited the proliferation of Mc3 cells in a dose-dependent and time-dependent manner. As the concentration increased, ADS I has exerted significant anti-proliferative effect on Mc3 cell. The IC50 value of ardipusilloside I was 9.98 ug/ ml. The apoptosis rates in 2.5,5,10ug/ml groups were 15.57±0.07%,25.63±0.18% and 86.27±0.04%respectively, much higher than that in control group (11.63±0.16%). Microscopy demonstrated apoptotic characteristics, including the nuclear chromatin condensation and cell nucleus fragmentation, as well as the ladder pattern revealed by agarose gel electrophoresis of DNA in a concentration-dependent manner. ADS I exposure also gradually increased the G0-G1 fraction and an S phase-arrest of both Mc3 cells. Furthermore, western-blot staining showed ADS I decreased the expression of Bcl-2 protein and increased the expression of caspase-3 and Bax protein.Conclusions:1. ADS I possess high inhibiting activity to proliferation of MC3 cells and can induce the cell cycle arrest. The cell proliferation is inhibited by ADS I in dose and time dependent manners.2. At the approximate the concentration, ADS I can induce apoptosis of Mc3 cells significantly.3.
