Protection Of Radix Astragali Injection On The Peripheral Neuronal Mitochondria Of Intracerebral Hemorrhage Foci In Rats

Objective To study the protective effect of radix astragali injection on the peripheral neuronal mitochondria of intracerebral hemorrhage foci in rats.Method ICH model was established by injecting collagenaseⅦinto the brain of rats. 120 male rats were randomly divided into the radix astragali injection remedial group and the contrast groups including ICH contrast group and normal contrast group without ICH. Radix astragali injection remedial group of 24h before the operation and radix astragali injection remedial group of 6h and 24h after the operation were administrated with the dose 2ml/kg. Then the radix astragali injection remedial groups were divided into 3 groups which were radix astragali injection remedial group of 24h before the operation and radix astragali injection remedial group of 6h and 24h after the operation. All rats involved in the experiment were killed on the 2d,3d,7 d after ICH. To detect the ethology of rats with every group, the changes of cytochrome C, Na+-K+-ATPase and Ca2+-Mg2+ATPase activities, intracellular Ca2+ concentration, and mitochondrium transmembrance potential etc.Results The result of neural behavioral test,After recovery from anesthesia, Rats in each group suffered from symptoms of hemiplegia, Manifested as cerebral hemorrhage contralateral limbs to reduce or limb weakness, forelimb is the most serious. The neurological deficit in the majority of rats showed crawling or can not stand rotating, Active motion decrease, and so on. With the passage of time, the treated rats gradually reduce the neurological deficit. The expression of Cyt-C: compared with normal contrast group, the expression of Cyt-C increased obviously on the 2d,3d ICH contrast groups (P<0.01), but there was no change on the 7d ICH contrast group; Compared with ICH contrast group, the expressions of Cyt-C of 2d and 3d remedial groups before and after operation decreased obviously (2d-24h and 3d-24h groups P<0.05;2d and 3d remedial groups of pretreatment,2d-6h and 3d-6h groups (P<0.01), The expression of Cyt-C of 3d remedial group of pretreatment was lower than 3d-24h (P<0.05), there was no change among the other remedial groups (P>0.05). The change of the activity of mitochondrium ATPase: the activity of Na+-K+-ATPase and Ca2+-Mg2+ATPase on the ICH contrast group were lower obviously than normal contrast group (P<0.01). Compared with the ICH contrast group, the activities of remedial groups increased (P<0.05), Among the remedial groups, there were significant changes between 2d remedial group of pretreatment and 2d-24h remedial group, 2d-6h and 2d-24h group (P<0.05). The change of mitochondrium transmembrance potential: the fluor pixel value increased more on the ICH contrast group than on the normal contrast group (P<0.01); compared with ICH contrast group, remedial groups'decreased obviously (P<0.01). preventive medication groups were better than 2d,3d and 7d remedial groups (P<0.01), 2d6h was better than 2d-24h (P<0.05), 3d6h was better than 3d-24h (P<0.01), there were no change between 7d24h and 7d2h (P>0.05). The change of concentration of Ca2+, compared with normal contrast group, the concentration of Ca2+ increased; compared with ICH contrast groups, the [Ca2+] decreased obviously of remedial groups(P<0.01), moreover the remedial groups before operation were better than remedial groups after operation(P<0.01), 2d-6h remedial group was better than 2d-24h,3d-6h remedial group was better than 3d-24h(P<0.05). there were no change between 7d-24h and 7d-2h (P>0.05). Conclusion①The expression of CytC increased obviously on the intracerebral hemorrhage foci in the earlier period, and then gradually descended. The radix astragalia injection could inhibit the release of mitochondria CytC.②With the lapse of time, the radix astragalia injection could heighten the activity of ATP enzyme on the 7d after intracerebral hemorrhage, lessen the aggregation of calcium and alleviate the dissipation of mitochondria transmembrane potential.③The radix of intracerebral hemorrhage foci on every remedial group, furthermore the effect was better if it was used 24 h before the operation and accordingly inhibit the occurrence of neuronic apoptosis;④The radix astragalia injection might inhibit the conduction of earlier apoptosis signals of peripheral neurons on the intracerebral hemorrhage foci through the pathway of mitochondria.
