Objective To investigate the effect of the Astragalus injection and the Astragaloside IVon the neuronal apoptosis around the focal of intracerebral hemorrhage in rats and explore theregulation of neuronal apoptosis and apoptotic pathway after cerebral hemorrhage,in order toprovide expeimental basis for clinical application of Astragalus IV and Astragalus injection.Method140healthy adult SD rats were selected without limitation of sex and weighing300±50g. Rats were randomly divided into three groups: the normal group (n=20), modelgroup (n=40) and treatment group (n=80). Model group were divided into3d group (n=20)and7d group (n=20). Treatment group were divided into Ags injection3d group (n=20),7d group(n=20) and Ags IV3d group (n=20),7d group (n=20). VII collagenase were injected into therat caudate nucleus to create models of intracerebral hemorrhage by stereotaxic instrument.After the success of the ICH model was produced6h, the treatment groups rats were injectedAgs injection or Ags IV by intraperitoneal, and then every24h inject one time. Dailyintraperitoneal injection of the corresponding amount of saline to model group. Each group ofrats were separately conducted for Neurological behavior, Nissl staining, Giemsa staining,IHC staining, Brain water content and Western Blot experimental observations.Results①Rat neurobehavioral score: the Astragalus injection treatment group and theAstragaloside IV treatment group can effectively reduce the level of the neurological deficitscore of intracerebral hemorrhage in rats (P<0.05); The Astragalus injection and theAstragaloside IV treated the intracerebral hemorrhage in rats at7d neurological behavior toimprove the situation better than the corresponding3d treatment group (P<0.05).②The brain water content after intracerebral hemorrhage: Compared with normal group, the brain watercontent of intracerebral hemorrhage model group was significantly increased (P<0.05);Compared with the corresponding ICH model group, the brain water content of each treatmentgroup rats were significantly decreased (P<0.05); Compared with the each time of Ags IVtreatment group, the brain water content of the each time of Ags treatment group rats weresignificantly decreased (P<0.05); Compared with the each treatment groups of3d, the brainwater content of the each treatment groups of7d was significantly reduced (P<0.05). The brainwater content of7d Astragalus injection treatment group rats were near the normal level of rats.③Rat brain tissue Nissl staining: The Nissl bodies in normal neurons were large in size andnumber, stained dark; The Nissl bodies in light injury neuron were small in size and number,stained light. The number of Nissl bodies in serious injury neurons were fewer.④Rat braintissue Giemsa staining: Compared with model group, the number and extent of the neuronalapoptosis in perihematomal of the treatment groups was significantly decreased.⑤Immunohistochemical staining of rats brain tissue: In the normal brain tissue, there was only afew expression of the Bax, Bcl-2and Caspase-3, Bcl-2and Bax ratio values close to1.Compared with the corresponding ICH model group, Bcl-2protein expression of eachtreatment group was significantly increased (P<0.05), but Bax and Caspase-3proteinexpression were significantly reduced (P<0.05). Compared with the3d Ags IV treatmentgroup, Bcl-2protein expression of the3d Ags treatment group was significantly increased(P<0.05), Bax and Caspase-3protein expression were significantly reduced (P<0.05).Compared with the7d Ags IV treatment group, Bcl-2protein expression of the7d Agstreatment group was significantly increased (P<0.05), Bax and Caspase-3protein expressionwere significantly reduced (P<0.05). Compared with the3d Ags treatment group, Bcl-2protein expression of the7d Ags treatment group was significantly increased (P<0.05), Baxand Caspase-3protein expression were significantly reduced (P<0.05). Compared with the3dAgs IV treatment group, Bcl-2protein expression of the7d Ags IV treatment group was significantly increased (P<0.05), Bax and Caspase-3protein expression were significantlyreduced (P<0.05).⑥Rat brain tissue by Western blot: In the normal brain tissue, there wasonly a few expression of the Bax, Bcl-2and Caspase-3. Compared with the brain tissue of thenormal rats, the Bax、Bcl-2and Caspase-3protein expression of each ICH model group wereincreased, and the Bax and Caspase-3protein expression increased particularly significantly(P<0.05). Compared with the corresponding ICH model group, Bcl-2protein expression ofeach treatment group were significantly increased (P<0.05), the protein expression of Bax andcaspase-3were significantly reduced (P<0.05). Compared with the each treatment groups of3d, Bcl-2protein expression of the each treatment groups of7d was significantly increased(P<0.05), Bax and Caspase-3protein expression was significantly reduced (P<0.05).Compared with the each time of Ags IV treatment group, Bcl-2protein expression of the eachtime of Ags treatment group was significantly increased (P<0.05), Bax and Caspase-3proteinexpression was significantly reduced (P<0.05).Conclusion①Astragaloside IV and Astragalus injection could extent to promote theabsorption of brain edema after ICH, can promote the recovery of neurological defects inintracerebral hemorrhage.②The Astragaloside IV and Astragalus injection can minimize theperihematomal the nerve cells apoptosis. Compared with Astragaloside IV, Astragalusinjection on perihematomal decreased expression of apoptotic cells is more obvious.
