Objective To investigate the effect of heat shock protein 70 induced by alanyl-glutamine on inhibiting tubulointerstitial fibrosis in unilateral ureteral obstruction rats.Method 20 male wistar rats weighing 200~250g were randomly divided into four groups:sham group,UUO model group,GLN group and QUE group. Rats subjected to UUO operation were treated with daily injection dose of 1.125g/kg Ala-Gln or 0.9% NS one day before UUO operation until being sacrificed.Rats of QUE group were treated with daily injection dose of 400mg/kg quercetin one day before injection of Ala-Gln. Five rats were sacrificed at day 7 in each group.Routine kidney pathology and Masson assay was examined.Expression of HSP70,NF-κB and a-SMA protein was assessed by immunofluorescence and Western-blot.Results GLN group markedly elevated HSP70 level in kidneys.As compared to UUO model group and QUE group,GLN group significantly reduced tubular lesions and attenuated interstitial fibrosis [ ( 26.2%±1.6% vs 78.4%±3.5%,80.2%±3.3%),score(1.32±0.08 vs3.67±0.22,3.70±0.20),P <0.05]; TEMT was significantly reduced by down-regulation of NF-κB and a-SMA protein (P<0.05)Conclusion Ala-Gln can markedly attenuate tubulointerstitial fibrosis in UUO rat model by up-regulation of HSP70 in kidneys ,and the inhibition of TEMT and a-SMA by down-regulation of NF-κB is possibly one of the potential mechanisms.
