Objective To investigate the effects of alanyl-glutamine on lipopoly- saccharide induced acute lung injury in rat and its mechanisms. Methods Sixty healthy adult Wistar rats were randomly divided into 6 groups of 10 each: saline control group(C); LPS group (LPS); alanyl-glutamine group (A1); glutamine group (G1); LPS and alanyl-glutamine group(1 hour after inject LPS)(A2); LPS and glutamine group(1 hour after inject LPS )(G2). Survival rate was observed after six hours of administration of LPS. Blood was drawn for analysis of eytokine levels before inject(T0) and 6h after administration of LPS (T1), all rats were killed after 6h hours of administration of LPS. The serum TNF-α,IL-1β,IL-8 were detected by ELISA , the lung wet/dry weight ratio and the content of protein in the bronchoalveolar lavage fluid(BALF) were determined. The extent and location of cell apoptosis in the bronchus and lung tissues were studied by TUNEL technique. The expression of HSP70 was detected by western blot. Results Survival rate after six hours of administration of LPS both are 70% in group A2,G2. Six hours after injection of LPS (T1), the lung wet/dry weight ratio, the content of protein in the BALF and the level of TNF-α,IL-1β,IL-8 was significantly lower in group A1,G1 than that in group LPS(P<0.05). The apoptosis index of both alveolar and bronchial epithelial cells was significantly lower in A1,G1 group than that in LPS group(P<0.05). Gray gradients of HSP70 in lungs in A1,G1 group was significantly higher than those of group LPS ( P < 0.05 ) . Conclusion Preadministration of alanyl-glutamine enhanced HSP70 expression significantly, and markedly reduced the serum levels of TNF-α,IL-1β,IL-8, inhibited...
